小分子物質(zhì)和牛血清白蛋白的作用及納米材料對(duì)其影響方面的一些研究

【摘要】:血清白蛋白(SA)是人體中一種十分重要的蛋白質(zhì),它有許多功能,包括和藥物小分子的吸收結(jié)合、與食品中有害小分子的作用等。納米材料的特有尺寸,使其相對(duì)于其它材料有很多優(yōu)異的性能,并在許多領(lǐng)域,包括化學(xué)、醫(yī)學(xué)等得到廣泛的應(yīng)用。而將納米材料用作某些藥物的載體是研究比較前沿和科研工作者感興趣的課題。本論文從分子水平上研究了幾種常見(jiàn)的小分子,包括抗癌藥物白藜蘆醇、煙葉中有害小分子尼古丁以及常作為補(bǔ)鐵劑的氯化血紅素與牛血清白蛋白(BSA)的作用,通過(guò)熒光光譜法等測(cè)試手段獲得這幾種小分子物質(zhì)與BSA相作用的信息。我們還在該作用過(guò)程中引入納米粒子,考察納米粒子對(duì)該反應(yīng)可能產(chǎn)生的影響,以對(duì)納米粒子的應(yīng)用作出一些有益的結(jié)論。本文的研究由三個(gè)方面組成: 1.研究了煙草中主要致煙癮物質(zhì)尼古丁與BSA二者的反應(yīng)情況,并在尼古丁和BSA的反應(yīng)過(guò)程中加入銀納米粒子(Ag NPs),將得到的結(jié)果與之前未加入Ag NPs時(shí)的結(jié)果進(jìn)行比較,可以看到在體系中加入Ag NPs后會(huì)抑制尼古丁與BSA的結(jié)合。進(jìn)而考察了Ag NPs抑制BSA與尼古丁結(jié)合的原因,運(yùn)用幾種光譜學(xué)方法來(lái)研究BSA與Ag NPs的結(jié)合,結(jié)果表明Ag NPs與BSA的結(jié)合會(huì)引發(fā)BSA二級(jí)結(jié)構(gòu)的改變,改變了尼古丁和BSA的結(jié)合狀態(tài)。 2.使用了熒光光譜、紫外吸收光譜以及微分脈沖伏安法研究補(bǔ)鐵劑氯化血紅素和BSA的作用,將得出的結(jié)果與體系中同時(shí)存在單寧酸的情況進(jìn)行對(duì)比,可以看到在實(shí)驗(yàn)體系中加入單寧酸后,氯化血紅素和BSA的結(jié)合變?nèi)?即結(jié)合常數(shù)變小,以及結(jié)合位點(diǎn)數(shù)減小, BSA二級(jí)結(jié)構(gòu)的改變程度也變得更弱。另一方面還研究了單寧酸對(duì)氯化血紅素和BSA之間結(jié)合力的改變情況,得出單寧酸的存在并不能改變氯化血紅素和BSA的結(jié)合力類(lèi)型的結(jié)論。 3.首先根據(jù)文獻(xiàn)合成聚乙二醇包裹的四氧化三鐵納米粒子(PEG-Fe3O4),并進(jìn)行表征,運(yùn)用熒光和紫外光譜法研究PEG-Fe3O4和抗癌藥物白藜蘆醇的吸附作用,結(jié)果顯示納米粒子會(huì)和白藜蘆醇作用進(jìn)而求得納米粒子對(duì)白藜蘆醇的載藥量,隨后研究了合成材料與BSA的相互作用,最后運(yùn)用光譜法結(jié)合化學(xué)計(jì)量學(xué)研究了白藜蘆醇與BSA的作用并考察了PEG-Fe3O4的存在對(duì)該結(jié)合作用的影響,驗(yàn)證了PEG-Fe3O4可以用作藥物載體的可能性。
[Abstract]:Serum albumin (SA) is an important protein in the human body, it has many functions, including small molecules and drug absorption combined with food harmful to the interaction of small molecules. The unique size of nano materials, compared with the other materials have many excellent properties, and in many fields including chemistry. Medicine is widely used. And nano materials will be used as a carrier of certain drugs is the comparative study of frontiers and researchers interested in the topic. This thesis studied several common small molecules at the molecular level, including anticancer drug resveratrol, small molecule nicotine and often used as iron supplement heme chloride and bovine serum albumin harmful tobacco (BSA) role, this was the role of several small molecules and BSA phase information by means of fluorescence spectroscopy test. The process we are introducing nanoparticles The effects of nanoparticles on the reaction are investigated in order to make some useful conclusions on the application of nanoparticles. The study in this paper is composed of three aspects:
1. the study of the reaction of tobacco smoking material mainly caused by nicotine and BSA two, and joined the silver nanoparticles in the reaction process of nicotine and BSA (Ag NPs), comparing the results obtained with before joining Ag NPs results, combined with the addition of Ag NPs can be seen in the system will inhibit nicotine with BSA. In order to investigate the cause of Ag NPs inhibition of BSA in combination with nicotine, combined with BSA and Ag NPs to study the use of several spectroscopic methods, the results show that the combination of Ag NPs and BSA BSA two will lead to changes in the structure, change the binding state of nicotine and BSA.
2. using fluorescence spectroscopy, UV absorption spectroscopy and differential pulse voltammetry to study effects of iron supplement hemin and BSA, the results and the system exist at the same time compared with that of the tannic acid, tannic acid added in the experiment can be seen in the system after the heme chloride and BSA binding is weak, binding constant becomes small, and the number of binding sites decreased, change degree of BSA two level structure has become more weak. On the other hand also studied the tannic acid on the binding force between hemin and BSA changes, the presence of tannins the type of binding force does not change the conclusion of hemin and BSA.
3. according to the fe3o4nanoparticles synthesis of polyethylene glycol (PEG-Fe3O4), the package and adsorption were characterized by fluorescence and UV spectroscopy of PEG-Fe3O4 and anticancer drug resveratrol, results showed that drug loading nanoparticles and effect of resveratrol and resveratrol nanoparticles obtained, then study the interaction of synthetic materials and BSA, the last of resveratrol and BSA combined with chemometrics using spectral method and the effects of PEG-Fe3O4 on the existence with the effect, verify the possibility of PEG-Fe3O4 can be used as a drug carrier.
