EB病毒BARF1原型和變異型基因對鼻咽癌細胞生物學行為的影響

【摘要】:背景和目的:EB病毒(Epstein-Barr virus, EBV)編碼早期基因BARF1是新近確立的EBV致癌基因之一,其轉化作用已在多種細胞系中得到證實。由于BARF1在多數鼻咽癌(nasopharyngeal carcinoma, NPC)和EBV相關胃癌組織中表達,推測其可能在NPC、胃癌等上皮細胞性腫瘤的發(fā)生中發(fā)揮重要作用。我們前期在檢測BARF1基因多態(tài)性時發(fā)現(xiàn)V29A變異型在NPC中的檢出率高于健康人群。本研究旨在探討B(tài)ARF1基因對NPC細胞系腫瘤原性的影響以及BARF1基因變異型對其致癌潛能的影響,為研究BARF1參與NPC的作用及機制提供依據。 方法:構建BARF1基因原型(B95-8)和變異型(V29A)慢病毒表達載體,分別轉染EBV陰性NPC細胞系CNE,殺稻瘟菌素篩選,RT-PCR和qRT-PCR鑒定目的基因表達獲得BARF1原型和變異型基因穩(wěn)定轉染細胞。以對照病毒轉染細胞為對照,進行MTT實驗、細胞周期檢測、平板克隆形成實驗、凋亡誘導實驗和細胞遷移實驗,檢測BARF1原型和變異型基因對CNE細胞生物學行為的影響。 結果:(1)成功構建BARF1基因原型和變異型基因穩(wěn)定轉染細胞,2種基因在EBV陰性NPC細胞系中均能有效表達。(2)細胞增殖實驗:BARF1原型和變異型基因轉染細胞及對照病毒轉染細胞MTT吸光度值、平板克隆形成率和細胞周期細胞增殖指數均無顯著性差異(P均大于0.05)。(3)凋亡誘導實驗:順鉑(2μg/ml)誘導48h,感染BARF1原型和變異型慢病毒細胞的存活率均高于對照病毒感染細胞,差異有顯著性(P均小于0.05),而細胞凋亡率低于對照病毒感染細胞,差異有顯著性(P均小于0.05),變異型與原型之間細胞存活率和凋亡率均無顯著性差異(P均大于0.05)。(4)細胞遷移實驗:BARF1原型和變異型慢病毒感染細胞的遷移能力高于對照病毒感染細胞,差異有顯著性(P0.05),變異型與原型之間細胞遷移無顯著性差異(P0.05)。 結論:(1)BARF1基因對EBV陰性NPC細胞系CNE無明顯促增殖作用,但可提高其抗凋亡能力和遷移能力,從而增強CNE細胞腫瘤原性,在NPC發(fā)生發(fā)展過程中可能起重要作用。(2)BARF1V29A變異型對BARF1致癌潛能無明顯影響。
