PTTG和VEGF-C在喉癌組織中的表達(dá)及其與微淋巴管生成的關(guān)系

【摘要】:目的探討垂體腫瘤轉(zhuǎn)化基因(PTTG)和血管內(nèi)皮生長因子-C(VEGF-C)在喉癌組織中表達(dá)的相關(guān)性以及與喉癌發(fā)展的關(guān)系。方法應(yīng)用免疫組織化學(xué)SP法分別檢測60例喉癌組織和32例癌旁組織中PTTG和VEGF-C兩種蛋白的表達(dá)情況,并用D2-40陽性產(chǎn)物定位淋巴管內(nèi)皮細(xì)胞細(xì)胞質(zhì)及細(xì)胞膜,計(jì)數(shù)微淋巴管密度(LMVD)。結(jié)果喉癌組織中PTTG和VEGF-C表達(dá)水平均顯著高于癌旁組織,差異有統(tǒng)計(jì)學(xué)意義(P0.05);二者表達(dá)量與喉癌的分化程度、有無淋巴結(jié)轉(zhuǎn)移及臨床分期差異均有統(tǒng)計(jì)學(xué)意義(P0.05),與臨床分型、有無吸煙史、腫瘤大小、性別及年齡方面的差異均無統(tǒng)計(jì)學(xué)意義(P0.05)。喉癌組織中LMVD表達(dá)水平明顯高于癌旁組織,差異有統(tǒng)計(jì)學(xué)意義(P0.05),與淋巴結(jié)轉(zhuǎn)移顯著相關(guān)(P0.05),與分化程度、臨床分期、臨床分型、有無吸煙史、腫瘤大小、性別及年齡不相關(guān)(P0.05)。PTTG和VEGF-C在喉癌組織中表達(dá)呈顯著正相關(guān)(P0.05)。LMVD分別與喉鱗癌組織中PTTG和VEGF-C蛋白表達(dá)正相關(guān)(P0.05)。結(jié)論 PTTG和VEGF-C在喉癌的發(fā)展中起重要作用,二者的檢測可作為喉癌轉(zhuǎn)移及預(yù)后的預(yù)判指標(biāo),有望成為新的基因治療靶點(diǎn)。
[Abstract]:Aim to investigate the relationship between the expression of pituitary tumor transforming gene (PTTG) and vascular endothelial growth factor-C (VEGF-C) in laryngeal carcinoma and its relationship with the development of laryngeal carcinoma. Methods Immunohistochemical SP method was used to detect the expression of PTTG and VEGF-C proteins in 60 cases of laryngeal carcinoma and 32 cases of paracancerous tissues. Counting microlymphatic vessel density (LMVD). Results the expression levels of PTTG and VEGF-C in laryngeal carcinoma tissues were significantly higher than those in paracancerous tissues (P0.05). There was significant difference between the expression level and differentiation degree of laryngeal carcinoma, lymph node metastasis and clinical stage (P0.05), as well as clinical classification, smoking history, tumor size, and so on. There was no significant difference in sex and age (P0.05). The expression level of LMVD in laryngeal carcinoma was significantly higher than that in paracancerous tissues (P0.05), and correlated with lymph node metastasis (P0.05), differentiation degree, clinical stage, clinical classification, smoking history, tumor size, and so on. There was a significant positive correlation between the expression of PTTG and VEGF-C in laryngeal carcinoma (P0.05). LMVD was positively correlated with the expression of PTTG and VEGF-C protein in laryngeal squamous cell carcinoma (P0.05). Conclusion PTTG and VEGF-C play an important role in the development of laryngeal carcinoma. The detection of them can be used as predictors of metastasis and prognosis of laryngeal carcinoma and may be a new target of gene therapy.
