【摘要】：目的探討姜黃素與5氟尿嘧啶(5-FU)聯(lián)合應(yīng)用對鼻咽癌CNE-2Z細(xì)胞的生長抑制作用及誘導(dǎo)凋亡作用,并研究其機(jī)制。方法姜黃素、5-FU單獨(dú)應(yīng)用及兩者聯(lián)合應(yīng)用作用于鼻咽癌CNE-2Z細(xì)胞72 h,MTT法檢測細(xì)胞生長狀況,流式細(xì)胞儀檢測鼻咽癌細(xì)胞的凋亡率,Western blot檢測Bcl-2、Bax、caspase-3以及NF-κB蛋白表達(dá)水平變化。Real-time PCR檢測caspase家族中caspase-3、caspase-7、caspase-9 mRNA表達(dá)水平的變化。結(jié)果凋亡率檢測結(jié)果顯示:兩藥聯(lián)合應(yīng)用后,腫瘤細(xì)胞的凋亡率明顯高于單用藥組(P0.01)。姜黃素組、5-FU組和藥物合用組CNE-2Z細(xì)胞的Bcl-2蛋白、細(xì)胞核內(nèi)及細(xì)胞漿內(nèi)NF-κB蛋白的表達(dá)水平下降,Bax蛋白、Cleaved caspase-3蛋白的表達(dá)水平增高。聯(lián)合用藥組caspase-3、caspase-7和caspase-9 mRNA表達(dá)水平高于單用藥組。結(jié)論姜黃素和5-FU抑制鼻咽癌CNE-2Z細(xì)胞生長的作用是通過誘導(dǎo)細(xì)胞凋亡來實(shí)現(xiàn)的。藥物聯(lián)合應(yīng)用抑制鼻咽癌CNE-2Z細(xì)胞生長的作用可能是通過抑制Bcl-2基因的表達(dá),增強(qiáng)Bax基因的表達(dá),并通過抑制NF-κB活性來誘導(dǎo)凋亡而實(shí)現(xiàn)的。
[Abstract]:Aim to investigate the inhibitory effect of curcumin combined with 5-fluorouracil (5-FU) on the growth and apoptosis of nasopharyngeal carcinoma (NPC) CNE-2Z cells and its mechanism. Methods Nasopharyngeal carcinoma CNE-2Z cells were treated with curcumin and 5-FU alone or in combination for 72 hours. The growth of NPC cells was detected by MTT assay, and the apoptosis rate of NPC cells by, Western blot was detected by flow cytometry. Bcl-2,Bax, was detected by flow cytometry. The expression level of caspase-3 and NF- 魏 B protein was changed, and the expression level of caspase-3,caspase-7,caspase-9 mRNA in caspase family was detected by Real-time PCR. Results the results showed that the apoptosis rate of tumor cells was significantly higher than that of single drug group (P0.01). In curcumin group, 5-FU group and drug combination group, the expression of Bcl-2 protein, NF- kappa B protein in nucleus and cytoplasm of CNE-2Z cells decreased, while the expression level of Bax protein and Cleaved caspase-3 protein increased. The expression levels of caspase-3,caspase-7 and caspase-9 mRNA in the combined drug group were higher than those in the single drug group. Conclusion curcumin and 5-FU can inhibit the growth of nasopharyngeal carcinoma CNE-2Z cells by inducing apoptosis. The inhibitory effect of combination of drugs on the growth of nasopharyngeal carcinoma CNE-2Z cells may be achieved by inhibiting the expression of Bcl-2 gene and enhancing the expression of Bax gene, and inducing apoptosis by inhibiting the activity of NF- kappa B.
