【摘要】：利用不同濃度利多卡因(lidocaine)處理體外培養(yǎng)的人角膜上皮(HCEP)細(xì)胞系細(xì)胞,利用光鏡觀察、MTT、熒光染色、DNA電泳、TUNEL、流式細(xì)胞儀和透射電鏡方法研究了利多卡因?qū)CEP細(xì)胞的毒性作用及其機(jī)理。光鏡觀察和MTT檢測(cè)結(jié)果顯示,質(zhì)量濃度1.25~10.00 g/L的利多卡因?qū)CEP細(xì)胞具有顯著的毒性作用,并具有濃度和時(shí)間依賴性;AO/EB熒光雙染色結(jié)果顯示,質(zhì)量濃度0.625~10.000 g/L的利多卡因可引起HCEP細(xì)胞的質(zhì)膜通透性顯著提高,細(xì)胞凋亡率也具有濃度和時(shí)間依賴性;DNA電泳和TUNEL檢測(cè)結(jié)果顯示,利多卡因能引起HCEP細(xì)胞發(fā)生DNA斷片化;TEM觀察結(jié)果顯示,利多卡因能引起HCEP細(xì)胞的超微結(jié)構(gòu)出現(xiàn)了凋亡細(xì)胞的形態(tài)結(jié)構(gòu)特征,如胞質(zhì)空泡化、染色質(zhì)濃縮、線粒體膨脹且嵴的結(jié)構(gòu)紊亂、出現(xiàn)凋亡小體等;Annexin V/PI染色的流式細(xì)胞儀檢測(cè)結(jié)果顯示,利多卡因能引起HCEP細(xì)胞質(zhì)膜中的磷脂酰絲氨酸(PS)發(fā)生外翻變化;ELISA檢測(cè)結(jié)果顯示,利多卡因還能引起HCEP細(xì)胞中胱冬肽酶-3、-8、-9、-10表達(dá)量的增加,表明利多卡因確能引起HCEP細(xì)胞發(fā)生細(xì)胞凋亡,而不是細(xì)胞壞死。由此可見(jiàn),利多卡因在質(zhì)量濃度大于0.625 g/L時(shí)對(duì)HCEP細(xì)胞具有顯著的細(xì)胞毒性,并具有濃度和時(shí)間依賴性,且其毒性作用的發(fā)揮是通過(guò)誘導(dǎo)細(xì)胞凋亡實(shí)現(xiàn)的,在眼科臨床應(yīng)用中具有很大的毒副作用,應(yīng)謹(jǐn)慎使用。
[Abstract]:Human corneal epithelial (HCEP) cells were treated with lidocaine (lidocaine) at different concentrations. The cells were observed by light microscopy, MTT, fluorescence staining, DNA electrophoresis, TUNEL,. The toxic effect of lidocaine on HCEP cells and its mechanism were studied by flow cytometry and transmission electron microscopy. The results of light microscopy and MTT showed that lidocaine at 1.25g / L of 10.00 g / L had a significant toxic effect on HCEP cells in a dose-and time-dependent manner. The results of AO/EB fluorescence double staining showed that lidocaine (0.625 渭 g / L) could significantly increase the plasma membrane permeability of HCEP cells, and the apoptosis rate was also concentration-dependent and time-dependent. DNA electrophoresis and TUNEL analysis showed that lidocaine could induce DNA fragmentation in HCEP cells. The results of TEM showed that lidocaine could induce the ultrastructure of HCEP cells, such as cytoplasmic vacuolation, chromatin concentration, mitochondria swelling and structural disorder of crest, apoptotic corpuscles, etc. The results of flow cytometry with Annexin V/PI staining showed that lidocaine could induce the evagination of phosphatidyl serine (PS) in the cytoplasmic membrane of HCEP. The results of ELISA showed that lidocaine could also increase the expression of cysteinase-3-8 and 9-10 in HCEP cells, suggesting that lidocaine could induce apoptosis rather than necrosis of HCEP cells. It can be seen that lidocaine has significant cytotoxicity to HCEP cells when the mass concentration is greater than 0.625 g / L, and has a concentration-and time-dependent effect, and its toxic effect is achieved by inducing apoptosis. It has great toxicity and side effects in clinical application of ophthalmology, and should be used with caution.
