α-硫辛酸對(duì)小鼠視網(wǎng)膜光化學(xué)損傷的保護(hù)作用

【摘要】:目的觀察α-硫辛酸對(duì)光損傷后小鼠視網(wǎng)膜的保護(hù)作用。方法將42只成年健康BALB/C小鼠隨機(jī)分為正常組、光損傷組和藥物干預(yù)組(干預(yù)組又分為造模前后均給藥組及造模后給藥組)。除正常組外其他組小鼠均暴露于裝有白色光源的光損傷箱內(nèi),光暴露時(shí)間為12h,之后暗室飼養(yǎng)12h,共3個(gè)循環(huán),光照強(qiáng)度21400±1600 lx,總暴露時(shí)間為36h。箱內(nèi)溫度控制在25-29℃。藥物干預(yù)組腹腔注射α-硫辛酸注射液100mg/Kg,一次/日,直至處死。分別于7d及14 d處死動(dòng)物摘取眼球,制備眼球壁石蠟切片,在光學(xué)顯微鏡下觀察其形態(tài)學(xué)變化,,測(cè)量視網(wǎng)膜外核層厚度,用TUNEL法計(jì)數(shù)外核層細(xì)胞凋亡數(shù)及凋亡指數(shù),用x±s表示,各組之間比較用兩因素析因設(shè)計(jì)的方差分析。結(jié)果與光損傷組相比,藥物干預(yù)組視網(wǎng)膜結(jié)構(gòu)層次分明,細(xì)胞排列整齊。其中視網(wǎng)膜外核層厚度,正常組41.62±0.77um,光損傷組7天30.51±0.73um,14天20.04±0.52um,藥物干預(yù)組(C1組)7天40.52±0.80um,14天41.43±0.72um,造模后給藥組(C2組)7天31.70±0.0.70um,14天38.92±1.43um,顯著高于光損傷組,(p0.001)。Tunel試劑盒檢測(cè)細(xì)胞凋亡結(jié)果顯示:正常組AI1,光損傷組AI=15.44±1.72,藥物干預(yù)Ⅰ組AI=7.23±0.65,藥物干預(yù)Ⅱ組AI=7.48±0.88。給藥組小鼠視網(wǎng)膜外核層的凋亡細(xì)胞數(shù)明顯少于光損傷組小鼠。外核層(ONL)厚度及凋亡指數(shù)(AI)的組間差異具有統(tǒng)計(jì)學(xué)意義。結(jié)論在本實(shí)驗(yàn)條件下,α-硫辛酸對(duì)小鼠視網(wǎng)膜光損傷具有保護(hù)作用。
