p53、MDM2、eIF4E、EGFR在鼻咽癌組織中的表達及鼻咽癌臨床生物學行為相關(guān)性的分析

【摘要】:目的:通過對p53、MDM2、eIF4E及EGFR在正常鼻咽部粘膜、鼻咽部非典型增生和鼻咽癌組織中表達的比較,分析這些基因異常與鼻咽癌患者的年齡、性別、不同病理類型、淋巴結(jié)轉(zhuǎn)移等臨床病理參數(shù)是否存在相關(guān)性;各異;蛑g是否具有相關(guān)性,從而探討這些基因異常在鼻咽癌發(fā)生發(fā)展中的作用及其臨床意義。 方法:應用免疫組織化學兩步法染色方法,對97例的鼻咽癌(其中未分化非角化型15例,分化非角化型82例),鼻咽部非典型增生18例及正常健康成人鼻咽粘膜17例病理標本檢測p53、MDM2、eIF4E和EGFR的表達情況。所得數(shù)據(jù)采用PASW statistics 18.0的x2檢驗、Spearman相關(guān)分析進行統(tǒng)計處理。 結(jié)果:p53、MDM2、eIF4E、EGFR在鼻咽癌、不典型增生、正常鼻咽粘膜中的表達有顯著性差異(p<0.05)。p53、MDM2、eIF4E、EGFR在不同年齡分組、不同性別及不同T分期亞組中的表達無顯著性差異(p>0.05);MDM2與淋巴結(jié)轉(zhuǎn)移具有明顯相關(guān)性(p<0.05)。eIF4E在不同分化程度的鼻咽癌組織中的表達具有顯著差異性(p<0.05)。p53與MDM2的表達具有相關(guān)性(p<0.05)。 結(jié)論: 1.鼻咽癌組織、非典型增生組織及正常粘膜膜組織中p53、MDM2、eIF4E、EGFR蛋白陽性表達率均依次降低,具有顯著性差異,表明p53、MDM2、eIF4E、EGFR參與鼻咽癌的發(fā)生發(fā)展過程。 2.MDM2在鼻咽癌組織中的表達與淋巴結(jié)轉(zhuǎn)移具有相關(guān)性。 3.eIF4E的表達與鼻咽癌的分化程度具有相關(guān)性。 4.p53與MDM2在鼻咽癌中的表達具有明顯相關(guān)性,與EGFR、eIF4E無明顯相關(guān)性。
[Abstract]:Objective: to compare the expression of p53 MDM2eIF4E and EGFR in normal nasopharyngeal mucosa, atypical hyperplasia of nasopharynx and nasopharyngeal carcinoma (NPC), and to analyze the relationship between these gene abnormalities and age, sex and different pathological types of patients with nasopharyngeal carcinoma (NPC). The correlation of clinicopathological parameters such as lymph node metastasis; To explore the role of these abnormal genes in the development of nasopharyngeal carcinoma and their clinical significance. Methods: 97 cases of nasopharyngeal carcinoma (including 15 cases of undifferentiated non-keratinized type and 82 cases of differentiated non-keratinized type) were studied by immunohistochemical two-step method. The expression of p53 MDM2eIF4E and EGFR in 18 cases of nasopharyngeal atypical hyperplasia and 17 cases of normal nasopharyngeal mucosa were detected. The data were analyzed by PASW statistics 18. 0 x 2 test and Spearman correlation analysis. Results: the expression of EGFR in nasopharyngeal carcinoma, atypical hyperplasia and normal nasopharyngeal mucosa was significantly different (p < 0.05). There was no significant difference in expression between different sex and T stage subgroups (p > 0.05). There was a significant correlation between MDM2 and lymph node metastasis (p < 0. 05). The expression of eIF4E was significantly different in nasopharyngeal carcinoma with different differentiation degree (p < 0. 05). The expression of p53 was correlated with MDM2 expression (p < 0. 05). Conclusion: 1. The positive expression rate of p53 MDM2EIF4EEGFR protein in nasopharyngeal carcinoma, atypical hyperplasia and normal mucosal membrane decreased in turn, with significant difference, indicating that p53 MDM2eIF4EEGFR is involved in the genesis and development of nasopharyngeal carcinoma. The expression of 2.MDM2 in nasopharyngeal carcinoma is correlated with lymph node metastasis. The expression of 3.eIF4E was correlated with the degree of differentiation of nasopharyngeal carcinoma. The expression of 4.p53 and MDM2 in nasopharyngeal carcinoma was significantly correlated with EGFR,eIF4E.
