米諾環(huán)素對(duì)高眼壓模型視網(wǎng)膜功能的影響

【摘要】: 目的:探討米諾環(huán)素(minocycline)對(duì)兔慢性高眼壓引起的視網(wǎng)膜功能的改變,為該藥物在國(guó)內(nèi)應(yīng)用于眼科臨床治療視神經(jīng)損害性疾病提供實(shí)驗(yàn)研究依據(jù)。 方法:成年健康色素兔30只,隨機(jī)分為A、B、C三組,每組各10只。A組為空白對(duì)照組,B組為模型組,C組為米諾環(huán)素給藥組。選B、C組右眼作高眼壓模型,每眼抽取房水0.1ml,隨后注射0.3%復(fù)方卡波姆溶液0.1ml;C組給予米諾環(huán)素口服給藥,50mg*kg-1*d-1,首次加倍,連續(xù)30天。用Schiotz眼壓計(jì)測(cè)量并記錄眼壓,一周一次。30天時(shí),A、B、C組右眼均行光學(xué)相干眼底斷層掃描(OCT)、閃光全視野視網(wǎng)膜電圖(Flash electroretinogram, FERG)及閃光視誘發(fā)電位(Flash visual evoked potential, FVEP)檢查,通過光學(xué)相干眼底斷層掃描(OCT)檢測(cè)視神經(jīng)纖維層厚度,通過FERG觀察比較a波、b波振幅變化情況;通過FVEP觀察N1、P1、N2波峰時(shí)值的后延程度。最后用OLYMPUS光學(xué)顯微鏡在10×40倍鏡下觀察視網(wǎng)膜各層病理改變。SPSS13.0統(tǒng)計(jì)學(xué)分析軟件對(duì)結(jié)果進(jìn)行統(tǒng)計(jì)學(xué)分析。 結(jié)果:眼壓值:在整個(gè)實(shí)驗(yàn)期間B、C組動(dòng)物的眼壓均高于22mmHg,最高達(dá)55mmHg,且在造模后各個(gè)時(shí)間點(diǎn)均高于A組。OCT所檢測(cè)視神經(jīng)纖維層厚度測(cè)量結(jié)果統(tǒng)計(jì)分析顯示:視神經(jīng)纖維層厚度B組最薄,C組次之,A組最厚,差異均具有統(tǒng)計(jì)學(xué)意義(P0.05);FERG所檢測(cè)結(jié)果統(tǒng)計(jì)分析顯示:a波振幅B、C組較A組無明顯下降,三組比較差異均無統(tǒng)計(jì)學(xué)意義(P0.05);b波振幅B、C組較A組均有下降,且B組較C組下降明顯,差異均具有統(tǒng)計(jì)學(xué)意義(P0.05); FVEP所檢測(cè)各波結(jié)果統(tǒng)計(jì)分析顯示:三個(gè)波的峰時(shí)值B、C組較A組均有延遲,且B組較C組延遲明顯,差異均具有統(tǒng)計(jì)學(xué)意義(P0.05);光鏡下B組視網(wǎng)膜組織各層結(jié)構(gòu)最紊亂,A組視網(wǎng)膜組織各層結(jié)構(gòu)最完整,與A組比較,C組視網(wǎng)膜組織各層結(jié)構(gòu)改變,但較B組改變輕微。 結(jié)論:米諾環(huán)素對(duì)兔慢性高眼壓引起的視網(wǎng)膜損傷有保護(hù)作用。
[Abstract]:Objective: to investigate the effects of minocycline (minocycline) on retinal function induced by chronic intraocular pressure (IOP) in rabbits, and to provide experimental evidence for the clinical application of minocycline in the treatment of optic nerve injurious diseases in China. Methods: thirty adult healthy pigmented rabbits were randomly divided into three groups: group A (n = 10), group B (model group) and group C (n = 10) treated with minocycline. The right eyes of group B were selected as the model of high intraocular pressure, 0.1 ml aqueous humor was extracted from each eye, and then 0.1 ml of compound carbomer solution was injected into group C for oral administration of minocycline, 50 mg 路kg ~ (-1) 路d ~ (-1), doubling for the first time for 30 days. Intraocular pressure (IOP) was measured and recorded by Schiotz intraocular pressure meter. At 30 days a week, all right eyes in group C underwent optical coherence tomography (OCT),) flash pan-field electroretinogram (Flash electroretinogram, FERG) and flash visual evoked potential (Flash visual evoked potential,). FVEP), optical coherence tomography (OCT) was used to detect the thickness of optic nerve fiber layer, and the amplitude of a wave and b wave were observed and compared by FERG. The degree of delay of N _ 1 P _ 1N _ 2 peak was observed by FVEP. Finally, the pathological changes of various layers of retina were observed with OLYMPUS optical microscope under 10 脳 40 times microscope. The results were statistically analyzed by SPSS13.0 statistical analysis software. Results: intraocular pressure (IOP): the intraocular pressure of group B C was higher than 22 mm Hg, and the highest was 55 mm Hg during the whole experiment. Statistical analysis of the thickness of optic nerve fiber layer measured by OCT showed that the thickness of optic nerve fiber layer was the thinnest in group B, followed by group C, and thickest in group A. The difference was statistically significant (P0.05). The statistical analysis of FERG results showed that the amplitude of a wave in group C was not significantly lower than that in group A, and there was no significant difference among the three groups (P0.05). The amplitude of B wave in group B was significantly lower than that in group A (P0.05), and that in group B was significantly lower than that in group C (P0.05). Statistical analysis of the results of each wave detected by FVEP showed that the peak time of three waves in group C was delayed than that in group A, and the delay in group B was significantly higher than that in group C (P0.05). Under light microscope, the structure of each layer of retina in group B was the most disordered, and that in group A was the most complete. Compared with group A, the structure of each layer of retina in group C changed slightly, but it was slightly different from that in group B. Conclusion: minocycline has protective effect on retinal injury induced by chronic intraocular pressure in rabbits.
