【摘要】：目的探討糖尿病(DM)大鼠和正常大鼠角膜上皮修復(fù)過(guò)程中緊密連接蛋白o(hù)ccludin重塑的特點(diǎn)。方法 90只SD大鼠隨機(jī)分為DM組和正常對(duì)照組(NC)各45只(DM組均高脂喂養(yǎng)),通過(guò)腹腔注射鏈脲佐菌素(STZ)誘導(dǎo)形成Ⅱ型DM大鼠模型,于損傷后0 h(未損傷),16、48、72、120 h(每組5只)處死大鼠取角膜。間接免疫熒光染色和Western blot法觀察occludin蛋白在損傷修復(fù)過(guò)程中的變化。結(jié)果DM大鼠角膜上皮損傷愈合率較NC組大鼠明顯延遲,DM組和NC組大鼠角膜上皮occludin蛋白在損傷后16、48 h表達(dá)差異有統(tǒng)計(jì)學(xué)意義(P0.05),在損傷后0、72、120 h表達(dá)差異無(wú)統(tǒng)計(jì)學(xué)意義。結(jié)論糖尿病可影響大鼠角膜上皮創(chuàng)傷修復(fù)過(guò)程中緊密連接蛋白o(hù)ccludin蛋白的表達(dá)。
