鼻咽癌淋巴結(jié)轉(zhuǎn)移相關(guān)分子蛋白質(zhì)組學(xué)技術(shù)篩選與鑒定

【摘要】:目的:比較2組不同程度頸部淋巴節(jié)結(jié)轉(zhuǎn)移的初診鼻咽癌組織在蛋白質(zhì)組水平上的表達(dá)差異。方法:將52例2010-06-01-2011-03-31中山大學(xué)腫瘤防治中心初診鼻咽癌患者,按照頸部淋巴結(jié)轉(zhuǎn)移程度分為A組(N0~N1)和B組(N2~N3),分別收集每組患者的腫瘤組織,提取總蛋白,進(jìn)行雙向凝膠電泳分離和質(zhì)譜分析,利用UMAX PowerLook1100透射掃描儀獲取凝膠圖像,應(yīng)用PDQuest 7.1.0軟件包進(jìn)行圖像分析,選取凝膠圖譜中差異表達(dá)1.5倍且P0.01的蛋白質(zhì)斑點(diǎn),應(yīng)用基質(zhì)輔助激光解吸/電離-飛行時(shí)間質(zhì)譜儀(matrix assisted laser desorption ionization time of flight/mass spectrometry,MALDI-TOF/MS)進(jìn)行蛋白質(zhì)的肽指紋圖譜鑒定。結(jié)果:在2組頸部淋巴節(jié)結(jié)轉(zhuǎn)移程度不同的初診鼻咽癌患者腫瘤組織間,共有7個蛋白質(zhì)點(diǎn)強(qiáng)度差異≥1.5倍且P0.01。經(jīng)質(zhì)譜分析鑒定發(fā)現(xiàn),B組中的表達(dá)量比在A組中的表達(dá)量,Stathmin升高5倍,缺氧誘導(dǎo)因子1-α抑制劑升高10倍。結(jié)論:Stathmin和缺氧誘導(dǎo)因子1-α抑制劑在高淋巴結(jié)轉(zhuǎn)移的鼻咽癌組織中表達(dá)增加,提示其在腫瘤發(fā)生發(fā)展機(jī)制中的作用可能與淋巴結(jié)轉(zhuǎn)移有關(guān)。
[Abstract]:Aim: to compare the proteomic expression of nasopharyngeal carcinoma (NPC) with different degrees of cervical lymph node metastasis. Methods: 52 patients with nasopharyngeal carcinoma (NPC) at Cancer Prevention Center of Sun Yat-sen University, 2010-06-01-2011-03-31 were divided into two groups according to the degree of cervical lymph node metastasis: group A (N0~N1) and group B (N2~N3). The tumor tissues of each group were collected and the total protein was extracted. Two-dimensional gel electrophoresis separation and mass spectrometry analysis were performed. The gel images were obtained by UMAX PowerLook1100 transmission scanner and analyzed by PDQuest 7.1.0 software package. Protein spots with 1.5 times differential expression and P0.01 were selected. Matrix assisted laser desorption / ionization-time of flight mass spectrometer (matrix assisted laser desorption ionization time of flight/mass spectrometry,MALDI-TOF/MS) was used to identify the peptide fingerprint of protein. Results: in two groups of nasopharyngeal carcinoma patients with different degree of cervical lymph node metastasis, there were 7 protein spots with a intensity of more than 1.5 times and P0.01. The expression of Stathmin in group B was 5 times higher than that in group A, and the expression of hypoxia inducible factor 1- 偽 inhibitor was 10 times higher than that in group A. Conclusion: the expression of Stathmin and hypoxia inducible factor 1- 偽 inhibitor is increased in nasopharyngeal carcinoma with high lymph node metastasis, which suggests that the expression of Stathmin and hypoxia inducible factor 1- 偽 inhibitor may be related to lymph node metastasis.
