【摘要】：目的探討鋅指蛋白36(ZFP36)基因多態(tài)性與腫瘤壞死因子(TNF)-α及中重度阻塞性呼吸睡眠暫停綜合征(OSAS)的相關(guān)性。方法應(yīng)用TaqMan-PCR方法檢測(cè)2010年1月至2012年7月在新疆維吾爾自治區(qū)人民醫(yī)院高血壓科住院的408例中重度OSAS患者(OSAS組)和194名健康體檢者(對(duì)照組)的ZFP36 3個(gè)位點(diǎn)(rs251864、rs3746083及rs17879933)。采用放射免疫法檢測(cè)TNF-α。結(jié)果 OSAS組的頸圍、腹圍、體重指數(shù)、收縮壓、舒張壓、血尿酸、總膽固醇及TNF-α均明顯高于對(duì)照組(P0.05)。兩組在ZFP36 rs251864和rs17879933位點(diǎn)的基因型和等位基因頻率分布差異有統(tǒng)計(jì)學(xué)意義,TNF-α在上述2個(gè)位點(diǎn)的不同基因型差異有統(tǒng)計(jì)學(xué)意義(P0.05)。3個(gè)位點(diǎn)的不同基因型在呼吸睡眠相關(guān)指標(biāo)中,差異均無(wú)統(tǒng)計(jì)學(xué)意義。Logistic回歸分析結(jié)果顯示,TNF-α不是OSAS的獨(dú)立危險(xiǎn)因素。在控制了年齡、性別等混雜因素后,未發(fā)現(xiàn)ZFP36 3個(gè)位點(diǎn)基因多態(tài)性與OSAS之間存在相關(guān)性。結(jié)論中重度OSAS患者TNF-α明顯增高,同時(shí)ZFP36參與對(duì)TNF-α的調(diào)節(jié),但ZFP36 3個(gè)位點(diǎn)的突變不是中重度OSAS發(fā)生的獨(dú)立危險(xiǎn)因素。
[Abstract]:Objective to investigate the association of zinc finger protein 36 (ZFP36) gene polymorphism with tumor necrosis factor (TNF)-偽 and moderate to severe obstructive sleep apnea syndrome (OSAS). Methods from January 2010 to July 2012, the three ZFP36 loci (rs251864,rs3746083 and rs17879933) of 408 moderate and severe OSAS patients (OSAS group) and 194 healthy persons (control group) were detected by TaqMan-PCR method in hypertension department of Xinjiang Uygur Autonomous region people's Hospital from January 2010 to July 2012. TNF- 偽 was detected by radioimmunoassay. Results the neck circumference, abdominal circumference, body mass index, systolic blood pressure, diastolic blood pressure, uric acid, total cholesterol and TNF- 偽 in OSAS group were significantly higher than those in control group (P0.05). There were significant differences in genotype and allele frequency distribution between the two groups at ZFP36 rs251864 and rs17879933 loci. There were significant differences in the genotypes of TNF- 偽 between the above two loci (P0.05). Logistic regression analysis showed that TNF- 偽 was not an independent risk factor for OSAS. After controlling for age, sex and other confounding factors, there was no correlation between ZFP36 gene polymorphism and OSAS. Conclusion TNF- 偽 is significantly increased in moderate and severe OSAS patients, and ZFP36 is involved in the regulation of TNF- 偽, but the mutation of three ZFP36 loci is not an independent risk factor for the occurrence of moderate and severe OSAS.
