αB-晶狀體蛋白對大鼠急性高眼壓后視網(wǎng)膜神經(jīng)節(jié)細胞存活及凋亡蛋白Caspase-3表達的影響

【摘要】:目的 觀察αB-晶狀體蛋白對大鼠急性高眼壓后視網(wǎng)膜神經(jīng)節(jié)細胞存活及凋亡蛋白Caspase-3表達的影響。 方法 3月齡雄性Sprague-Dawley(SD)大鼠,體重250±20g ,160只完全隨機設(shè)計分成以下4組:A組(急性高眼壓組),B組(急性高眼壓+玻璃體穿刺組),C組(急性高眼壓+玻璃體注射生理鹽水組),D組(急性高眼壓+玻璃體注射αB-晶狀體蛋白組),應(yīng)用熒光金逆行標記計數(shù)法、免疫熒光法觀察急性高眼壓后各組大鼠視網(wǎng)膜神經(jīng)節(jié)細胞在不同時間點的存活率及凋亡蛋白Caspase-3表達情況。 結(jié)果 急性高眼壓后實驗動物右眼(實驗眼)視網(wǎng)膜神經(jīng)節(jié)細胞開始減少,低倍熒光顯微鏡下可見全視網(wǎng)膜節(jié)細胞均在減少,第一周減少最為明顯,兩周后減少放緩。與其它3組相比,D組在急性高眼壓后1周、2周、3周、4周視網(wǎng)膜神經(jīng)節(jié)細胞存活率明顯提高,差異具有統(tǒng)計學意義(P0.01),其它3組之間,差異無統(tǒng)計學意義(P0.05);急性高眼壓后實驗動物右眼(實驗眼)Caspase-3免疫熒光染色出現(xiàn)陽性表達,與其它3組相比,D組在急性高眼壓后24小時、48小時、72小時、1周的Caspase-3表達明顯減少,差異具有統(tǒng)計學意義(P0.01),其它3組之間,差異無統(tǒng)計學意義(P0.05)。 結(jié)論 αB-晶狀體蛋白對大鼠急性高眼壓后視網(wǎng)膜神經(jīng)節(jié)細胞存在保護作用,能增加視網(wǎng)膜神經(jīng)節(jié)細胞的存活率。 αB-晶狀體蛋白能抑制急性高眼壓后視網(wǎng)膜神經(jīng)節(jié)細胞內(nèi)凋亡蛋白Caspase-3的表達,減少細胞凋亡。
[Abstract]:objective
Objective To observe the effect of alpha B- crystallin on the survival of retinal ganglion cells and the expression of apoptosis protein Caspase-3 after acute ocular hypertension in rats.
Method
3 month old male Sprague-Dawley (SD) rats, weighing 250 + 20g, were divided into 4 groups randomly: group A (acute high intraocular pressure group), group B (acute ocular hypertension + vitreous puncture group), group C (acute ocular hypertension + vitreous injection of saline group), group D (acute ocular hypertension + vitreous injection of alpha B- lens protein group), using fluorescein retrograde labeling The survival rate of retinal ganglion cells and the expression of apoptotic protein Caspase-3 in different groups of rat retinal ganglion cells after acute ocular hypertension were observed by the method of counting and immunofluorescence.
Result
The retinal ganglion cells in the right eye (experimental eye) of the experimental animals began to decrease after acute intraocular pressure. Under the low fluorescence microscope, the retinal ganglion cells were reduced, the first week decreased most obviously and decreased after two weeks. Compared with the other 3 groups, the survival rate of retinal ganglion cells in the D group was 1 weeks, 2 weeks, 3 weeks and 4 weeks after acute ocular hypertension. The difference was statistically significant (P0.01), the difference between the other 3 groups was not statistically significant (P0.05). The positive expression of Caspase-3 immunofluorescence staining in the right eye (experimental eye) after acute ocular hypertension was positive. Compared with the other 3 groups, the expression of Caspase-3 in the D group was significantly reduced at 24 hours, 48 hours, 72 hours and 1 weeks after acute ocular hypertension. The difference was statistically significant (P0.01), and there was no significant difference between the other 3 groups (P0.05).
conclusion
Alpha B- crystallin has protective effect on retinal ganglion cells after acute ocular hypertension in rats, and can increase the survival rate of retinal ganglion cells.
Alpha B- crystallin can inhibit the expression of apoptosis protein Caspase-3 and reduce apoptosis in retinal ganglion cells after acute ocular hypertension.
