【摘要】：目的探討瘦素基因在甲狀腺相關(guān)眼病(thyroid associated ophthalmopathy,TAO)患者和正常人眼眶脂肪組織和體外誘導(dǎo)分化的眼眶脂肪細(xì)胞中的表達(dá)及意義。方法Trizol法提取8例(8眼)TAO患者和13例(13眼)正常人眼眶脂肪組織總RNA,采用實(shí)時熒光定量PCR檢測瘦素mRNA的表達(dá)。體外培養(yǎng)TAO患者與正常人眼眶前脂肪成纖維細(xì)胞并進(jìn)行誘導(dǎo)分化,實(shí)時熒光定量PCR檢測分化后脂肪細(xì)胞中瘦素mRNA的表達(dá)。結(jié)果 TAO患者眼眶脂肪組織中瘦素mRNA的相對表達(dá)量為2.05±0.31(已排除年齡、性別和身體質(zhì)量指數(shù)可能的影響),正常人眼眶脂肪組織中瘦素mRNA的相對表達(dá)量為0.69±0.24,兩者相比差異有統(tǒng)計(jì)學(xué)意義(P0.05)。體外培養(yǎng)的TAO患者和正常人眼眶前脂肪成纖維細(xì)胞在誘導(dǎo)分化刺激下,均能分化為脂肪細(xì)胞;TAO患者分化第10天眼眶脂肪細(xì)胞的瘦素mRNA的相對表達(dá)量為1.55±0.44,正常人分化第10天眼眶脂肪細(xì)胞的瘦素mRNA的相對表達(dá)量為0.66±0.21,兩者相比差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論瘦素基因在TAO患者眼眶脂肪組織和體外誘導(dǎo)分化的眼眶脂肪細(xì)胞中的表達(dá)均明顯高于正常人,提示瘦素基因可能參與了TAO的發(fā)病機(jī)制。
[Abstract]:Objective to investigate the expression and significance of leptin gene in adipose tissue of (thyroid associated ophthalmopathy (thyroid associated ophthalmopathy TAO) and normal human orbital adipocytes induced by thyroid associated ophthalmopathy in vitro. Methods Total RNAs were extracted from 8 patients (8 eyes) of TAO and 13 cases (13 eyes) of normal controls by Trizol method. The expression of leptin mRNA was detected by real-time fluorescence quantitative PCR. Preorbital adipose fibroblasts from TAO patients and normal subjects were cultured in vitro and induced to differentiate. The expression of leptin mRNA in differentiated adipocytes was detected by real-time fluorescence quantitative PCR. Results the relative expression of leptin mRNA in orbital adipose tissue of TAO patients was 2.05 鹵0.31 (excluded age). The relative expression of leptin mRNA in normal orbital adipose tissue was 0.69 鹵0.24, the difference was statistically significant (P0.05). Cultured preorbital adipose fibroblasts from TAO patients and normal subjects were induced and stimulated by differentiation. The relative expression of leptin mRNA in orbital adipocytes was 1.55 鹵0.44 on the 10th day of differentiation and 0.66 鹵0.21 in normal persons on the 10th day after differentiation. There was significant difference between the two groups (P0.05). Conclusion the expression of leptin gene in orbital adipose tissue of TAO patients and orbital adipocytes induced by differentiation in vitro is significantly higher than that in normal controls, suggesting that leptin gene may be involved in the pathogenesis of TAO.
