Cox2、Cx26、Stathmin在喉乳頭狀瘤組織中的表達(dá)及臨床意義

【摘要】: 目的通過(guò)檢測(cè)Cox2、Cx26及Stathmin蛋白在喉乳頭狀瘤及聲帶息肉組織中的表達(dá)情況,探討它們?cè)诤砣轭^狀瘤發(fā)生、發(fā)展過(guò)程中的作用及相互關(guān)系。 方法采用免疫組織化學(xué)技術(shù)(PV-9000二步法)檢測(cè)Cox2、Cx26及Stathmin蛋白在60例喉乳頭狀瘤和15例聲帶息肉組織中的表達(dá)情況,應(yīng)用Image-ProPlus圖像分析軟件測(cè)定免疫組化圖像的平均光密度(Mean Optical Density, MOD),運(yùn)用SPSS 13.0軟件進(jìn)行統(tǒng)計(jì)學(xué)分析,觀察各指標(biāo)與喉乳頭狀瘤臨床特征之間的關(guān)系及其在喉乳頭狀瘤組織中表達(dá)的相關(guān)性。 結(jié)果(1)Cox2及Stathmin蛋白在喉乳頭狀瘤組織中的陽(yáng)性表達(dá)率分別為73.33%、88.33%,均明顯高于它們?cè)诼晭⑷饨M織中的表達(dá)率(33.33%、53.33%),兩者差異有統(tǒng)計(jì)學(xué)意義(P0.05)。(2)Cx26蛋白在喉乳頭狀瘤組織中的陽(yáng)性表達(dá)率為33.33 %,明顯低于其在聲帶息肉組織中的表達(dá)率(73.33 %),兩者差異有統(tǒng)計(jì)學(xué)意義(P0.05)。喉乳頭狀瘤中Cx26蛋白的表達(dá)伴有細(xì)胞內(nèi)定位的改變,Cx26蛋白在聲帶息肉組織中主要表達(dá)在細(xì)胞膜,而在喉乳頭狀瘤組織中的細(xì)胞膜、細(xì)胞漿和小部分細(xì)胞核中均有陽(yáng)性表達(dá)。(3)Cox2、Cx26及Stathmin蛋白的表達(dá)水平與臨床分型、是否復(fù)發(fā)、患者性別及年齡均無(wú)相關(guān)性(P0.05)。(4)喉乳頭狀瘤中,Cox2蛋白與Stathmin蛋白表達(dá)呈正相關(guān)(r=0.287,P0.05),而Cx26蛋白表達(dá)與Cox2及Stathmin蛋白表達(dá)呈負(fù)相關(guān)(r=-0.213、r=-0.184,P0.05)。 結(jié)論 1. Cox2蛋白在喉乳頭狀瘤組織中高表達(dá),提示其與喉乳頭狀瘤的發(fā)生可能有密切關(guān)系,有望為選擇性Cox2抑制劑預(yù)防和治療喉乳頭狀瘤提供依據(jù)。 2.喉乳頭狀瘤組織中Cx26蛋白的表達(dá)明顯低于聲帶息肉,且伴有細(xì)胞內(nèi)定位的改變,提示其在喉乳頭狀瘤的發(fā)生、發(fā)展過(guò)程可能起重要的作用。 3. Stathmin蛋白在喉乳頭狀瘤組織中高表達(dá),提示其與喉乳頭狀瘤的多發(fā)性、浸潤(rùn)性可能有密切聯(lián)系,可能成為研究生物治療的方向之一。 4. Cox2蛋白與Stathmin蛋白在喉乳頭狀瘤組織中的表達(dá)呈正相關(guān),提示它們可能在喉乳頭狀瘤的發(fā)生、發(fā)展過(guò)程中起協(xié)同作用。Cx26蛋白與Cox2蛋白及Stathmin蛋白在喉乳頭狀瘤組織中的表達(dá)均呈負(fù)相關(guān),可能與它們之間的蛋白質(zhì)相互作用有關(guān),并可能與喉乳頭狀瘤的發(fā)生、發(fā)展有關(guān)。
[Abstract]:Objective to investigate the expression of Cox2Cx26 and Stathmin protein in laryngeal papilloma and vocal cord polyp, and to explore their roles in the development and development of laryngeal papilloma. Methods the expression of Cox2Cx26 and Stathmin protein in 60 cases of laryngeal papilloma and 15 cases of vocal cord polyps were detected by immunohistochemical technique (PV-9000 two-step method). The mean optical density (MOD) of immunohistochemical images was measured by Image-ProPlus image analysis software, and the statistical analysis was carried out by SPSS 13.0 software. To observe the relationship between the indexes and the clinical features of laryngeal papilloma and its expression in laryngeal papilloma. Results (1) the positive expression rates of Cox2 and stathmins in laryngeal papilloma were 73.33 and 88.33, respectively, which were significantly higher than those in vocal cord polyps (33.3353.33%). There was a significant difference between them (P0.05). (2) and Cx26 protein in laryngeal papilloma. The sexual expression rate was 33.33, which was significantly lower than that in vocal cord polyps (73.33%), the difference was statistically significant (P0.05). The expression of Cx26 protein in laryngeal papilloma is associated with the change of intracellular localization. Cx26 protein is mainly expressed in the cell membrane of vocal cord polyp, but in the cell membrane of laryngeal papilloma. (3) the expression level of Cox2Cx26 and Stathmin protein and clinical type, recurrence or not. There was no correlation between sex and age (P0.05). (4). There was a positive correlation between the expression of Cox2 protein and stathmin protein in laryngeal papilloma (P 0.05), but a negative correlation between the expression of Cx26 protein and the expression of Cox2 and stathmin protein (r-0.213r-0.184P0.05). Conclusion 1. The overexpression of Cox2 protein in laryngeal papilloma suggests that Cox2 protein may be closely related to the occurrence of laryngeal papilloma and may provide evidence for the prevention and treatment of laryngeal papilloma with selective Cox2 inhibitor. The expression of Cx26 protein in laryngeal papilloma was significantly lower than that in vocal cord polyp and accompanied by changes in intracellular localization, suggesting that Cx26 protein might play an important role in the occurrence and development of laryngeal papilloma. 3. The high expression of Stathmin protein in laryngeal papilloma suggests that Stathmin protein may be closely related to the multiple and invasive laryngeal papilloma and may be one of the research directions of biotherapy. Cox2 protein was positively correlated with the expression of stathmin protein in laryngeal papilloma, suggesting that Cox2 protein might occur in laryngeal papilloma. The expression of Cox2 protein and Stathmin protein in laryngeal papilloma was negatively correlated with the synergistic effect of Cx26 protein during the development of laryngeal papilloma, which may be related to the protein interaction between them, and to the occurrence and development of laryngeal papilloma.
