【摘要】：目的探討慢性扁桃體炎中人β-防御素-3(hBD-3)的表達(dá)及意義。方法應(yīng)用HE染色,逆轉(zhuǎn)錄聚合酶鏈?zhǔn)椒磻?yīng)、SP法、蛋白印跡法觀察和檢測(cè)15例扁桃體炎組和10例對(duì)照組組織的病理變化及組織中hBD-3的表達(dá)。結(jié)果 HE光鏡下顯示對(duì)照組病理特點(diǎn)固有層纖維結(jié)締組織反應(yīng)性增生為主,非角化復(fù)層扁平層內(nèi)未見(jiàn)炎性細(xì)胞浸潤(rùn);扁桃體炎組淋巴濾泡增生,生發(fā)中心擴(kuò)大,非角化復(fù)層扁平層內(nèi)炎性細(xì)胞浸潤(rùn),蛋白滲出明顯。逆轉(zhuǎn)錄聚合酶鏈?zhǔn)椒磻?yīng)結(jié)果顯示扁桃體炎組觀察到目的片段;對(duì)照組未見(jiàn)hBD-3的特異片段。免疫組織化學(xué)光鏡下顯示扁桃體炎組非角化復(fù)層扁平上皮細(xì)胞胞漿中hBD-3陽(yáng)性表達(dá);對(duì)照組非角化復(fù)層扁平上皮細(xì)胞胞漿中呈陰性表達(dá)。蛋白印跡法檢測(cè)結(jié)果顯示扁桃體炎組可見(jiàn)陽(yáng)性條帶;對(duì)照組未見(jiàn)陽(yáng)性條帶。結(jié)論 hBD-3在慢性扁桃炎中的表達(dá),表明hBD-3可能在扁桃體和上呼吸道抗感染中發(fā)揮重要作用。
[Abstract]:Objective to investigate the expression and significance of human 尾-defensin-3 hBD-3 in chronic tonsillitis. Methods the histopathological changes and the expression of hBD-3 in 15 cases of tonsillitis and 10 cases of control group were observed and detected by HE staining, reverse transcriptase polymerase chain reaction (RT-PCR) and Western blotting. Results under HE light microscope, the pathological features of the control group were fibrous connective tissue reactive hyperplasia in lamina propria, no inflammatory cell infiltration in the flat layer of non-keratinized lamina, lymphoid follicular hyperplasia and enlargement of germinal center in tonsillitis group. Inflammatory cells infiltrated in the flat layer of non-keratinized lamina, and protein exudation was obvious. Reverse transcriptase polymerase chain reaction showed that the target fragment was observed in tonsillitis group, but no specific hBD-3 fragment was found in control group. The expression of hBD-3 in cytoplasm of non-keratinized lamellar flat epithelial cells in tonsillitis group and control group was negative. The positive bands were found in tonsillitis group and control group. Conclusion the expression of hBD-3 in chronic tonsillitis suggests that hBD-3 may play an important role in anti-infection of tonsil and upper respiratory tract.
