HPA、CK2β和HIF-1α在鼻咽癌組織中的表達及意義

【摘要】:目的: 1、檢測乙酰肝素酶、蛋白激酶ck2β、缺氧誘導因子-1α在鼻咽癌組織中的表達以及與臨床特征之間的關(guān)系。 2、分析乙酰肝素酶、蛋白激酶ck2β、缺氧誘導因子-1α在鼻咽癌組織中表達的相關(guān)性。 3、探討乙酰肝素酶、蛋白激酶ck2β、缺氧誘導因子-1α的表達以及臨床特征與鼻咽癌患者預(yù)后的關(guān)系。 材料與方法: 采用免疫組織化學染色技術(shù)Super-Vision法檢測49例鼻咽癌(NPC)組織標本及15例慢性鼻咽炎(CNT)組織對照標本中HPA、CK2β及HIF-1α的表達,并應(yīng)用SPSS13.0統(tǒng)計軟件包進行相關(guān)臨床和病理參數(shù)分析。 結(jié)果: 1、HPA在NPC組織及CNT組織中的高表達率分別為49.0%與6.7%,差異有統(tǒng)計學意義(P=0.003)。HPA的表達與NPC臨床分期、治療后有無復(fù)發(fā)或轉(zhuǎn)移密切相關(guān),不同組別表達水平的差異均有統(tǒng)計學意義(P值分別為0.046、0.001)。HPA的表達與患者的性別、年齡無相關(guān)性,不同組別表達水平的差異均無統(tǒng)計學意義。 2、CK2β在NPC組織及CNT組織中的高表達率分別為44.9%與13.2%,差異有統(tǒng)計學意義(P=0.027)。CK2β的表達與NPC臨床分期、治療后有無復(fù)發(fā)或轉(zhuǎn)移密切相關(guān),不同組別表達水平差異均有統(tǒng)計學意義(P值分別為0.026、0.030)。CK2β的表達與患者的性別、年齡無相關(guān)性,不同組別表達水平的差異均無統(tǒng)計學意義。 3、HIF-1α在NPC組織及CNT組織中的高表達率分別為40.8與6.7%,差異有統(tǒng)計學意義(P=0.014)。HIF-1α的表達與NPC患者的年齡、臨床分期、治療后有無復(fù)發(fā)或轉(zhuǎn)移密切相關(guān),不同組別表達水平差異均有統(tǒng)計學意義(P值分別為0.002、0.027、0.005)。HIF-1α的表達與患者的性別無相關(guān)性,不同組別表達水平的差異無統(tǒng)計學意義。 4、HPA與CK2β、HPA與HIF-1α、CK2β與HIF-1α在NPC組織中的表達均呈正相關(guān)性(P值分別為0.012、0.006、0.000)。 5、NPC的臨床分期、治療后有無復(fù)發(fā)或轉(zhuǎn)移與患者的預(yù)后密切相關(guān),不同組別比較生存率的差異有統(tǒng)計學意義(P值分別為0.020、0.000)。HPA、CK2β、HIF-1α的表達水平與患者的預(yù)后密切相關(guān),不同組別比較生存率的差異有統(tǒng)計學意義(P值分別為0.001、0.019、0.011)。Cox回歸模型預(yù)后多因素分析顯示,僅治療后有無復(fù)發(fā)或轉(zhuǎn)移與患者的預(yù)后密切相關(guān),不同組別比較生存率的差異有統(tǒng)計學意義(P=0.001)。 結(jié)論: 1、HPA、CK2β和HIF-1α參與了NPC的生長、浸潤及轉(zhuǎn)移,HPA、CK2β和HIF-1α的高表達可促進NPC的發(fā)生和發(fā)展。 2、在NPC的發(fā)生發(fā)展過程中,HPA、CK2β和HIF-1α之間可能存在協(xié)同作用。 3、臨床分期、治療后有復(fù)發(fā)或轉(zhuǎn)移以及HPA、CK2β、HIF-1α的表達水平是影響患者預(yù)后的因素。 4、治療后有無復(fù)發(fā)或轉(zhuǎn)移是影響患者預(yù)后的獨立危險因素。
[Abstract]:Objective: 1. The expression of heparanase, protein kinase ck2 尾 and hypoxia inducible factor-1 偽 in nasopharyngeal carcinoma (NPC) and the relationship between heparanase, protein kinase ck2 尾 and clinical features were detected. 2. The correlation of heparanase, protein kinase ck2 尾 and hypoxia inducible factor-1 偽 expression in nasopharyngeal carcinoma was analyzed. 3. To investigate the relationship between the expression of heparanase, protein kinase ck2 尾, hypoxia inducible factor-1 偽 and the prognosis of nasopharyngeal carcinoma. Materials and methods: The expression of HPACK2 尾 and HIF-1 偽 in 49 cases of nasopharyngeal carcinoma (NPC) and 15 cases of chronic nasopharyngeal carcinoma (CNT) were detected by immunohistochemical staining Super-Vision method. The clinical and pathological parameters were analyzed by SPSS13.0 statistical software package. Results: 1the high expression rates of HPA in NPC and CNT were 49.0% and 6.7, respectively. The difference was statistically significant. The expression of HPA was closely related to the clinical stage of NPC and the recurrence or metastasis after treatment. There was no correlation between the expression level of HPA and sex and age, but there was no significant difference in expression level between different groups. 2the high expression rates of CK2 尾 in NPC and CNT were 44.9% and 13.2respectively. The difference was statistically significant. The expression of CK2 尾 was closely related to the clinical stage of NPC and the recurrence or metastasis after treatment. There was no correlation between the expression level of CK2 尾 and sex and age, but there was no significant difference in expression level between different groups. 3the high expression rates of HIF-1 偽 in NPC and CNT tissues were 40.8 and 6.7, respectively. The difference was statistically significant. The expression of HIF-1 偽 was closely related to the age, clinical stage, recurrence or metastasis of NPC patients. There was no correlation between the expression of HIF-1 偽 and the sex of the patients, but there was no significant difference in the expression level of HIF-1 偽 between different groups. (4) the expression of HPA and CK2 尾 -HPA and HIF-1 偽, CK2 尾 and HIF-1 偽 were positively correlated in NPC tissues (P = 0.012, P = 0.006, P = 0.000). 5The clinical stage, recurrence or metastasis after treatment were closely related to the prognosis of patients. The difference of survival rate in different groups was significantly different (P = 0.020 0.000). The expression level of HPACK2 尾 -HIF-1 偽 was closely related to the prognosis of patients. The difference of survival rate in different groups was statistically significant (P = 0.001 / 0.019 / 0.011.Cox regression model). Multivariate analysis showed that only recurrence or metastasis after treatment was closely related to the prognosis of the patients. The difference of survival rate in different groups was statistically significant. Conclusion: 1HPA-CK2 尾 and HIF-1 偽 participate in the growth of NPC, and the overexpression of HPA-CK2 尾 and HIF-1 偽 can promote the occurrence and development of NPC. 2. There may be a synergistic effect between HPACK2 尾 and HIF-1 偽 during the development of NPC. 3, clinical stage, recurrence or metastasis after treatment and the expression level of HPA-CK2 尾 -HIF-1 偽 were the prognostic factors. 4. Recurrence or metastasis after treatment is an independent risk factor for prognosis.
