重組人凋亡素2配體對(duì)放射線誘導(dǎo)鼻咽癌CNE細(xì)胞凋亡作用的研究

【摘要】:目的本實(shí)驗(yàn)旨在研究重組人凋亡素2配體(Apo-2ligand, Apo-2L)與放射線協(xié)同對(duì)體外培養(yǎng)的鼻咽癌CNE細(xì)胞增殖周期及凋亡率的影響。 方法MTT法檢測(cè)不同濃度Apo-2L對(duì)鼻咽癌CNE細(xì)胞凋亡的影響,并確定Aop-2L與細(xì)胞作用時(shí)間。將細(xì)胞隨機(jī)分為4組:對(duì)照組(CK組)、Apo-2L組(T組)、Apo-2L+放射組(T-R組)及單純放射組(R組);給予不同的組間處理后分別將4組細(xì)胞制成細(xì)胞懸液,將所得細(xì)胞懸液進(jìn)行流式細(xì)胞儀分析測(cè)得各組細(xì)胞凋亡率和細(xì)胞周期變化。 結(jié)果MTT法示鼻咽癌CNE細(xì)胞的抑制率與Apo-2L的濃度成正相關(guān);當(dāng)取Apo-2L濃度為100ng/ml作用CNE細(xì)胞24h,Apo-2L+放射組凋亡率較其它各組明顯升高,P0.05,有統(tǒng)計(jì)學(xué)意義;Apo-2L+放射組G2/M期細(xì)胞比率顯著降低,與其它各組相比有統(tǒng)計(jì)學(xué)差異,P0.05。 結(jié)論Apo-2L對(duì)體外培養(yǎng)的鼻咽癌CNE細(xì)胞有抑制增殖和促進(jìn)凋亡作用,其濃度與凋亡作用正相關(guān);Apo-2L聯(lián)合放射線可以明顯提高CNE細(xì)胞的凋亡率,并促使細(xì)胞周期同步化分布。
[Abstract]:Objective to investigate the effects of recombinant human apoptin 2 ligand Apo-2 ligand (Apo-2L) and radiation on the proliferation cycle and apoptosis rate of nasopharyngeal carcinoma (NPC) CNE cells cultured in vitro. Methods MTT assay was used to detect the effect of different concentrations of Apo-2L on the apoptosis of nasopharyngeal carcinoma (NPC) CNE cells, and to determine the time of interaction between Aop-2L and the cells. The cells were randomly divided into four groups: control group (CK group), Apo-2L group (T group) and simple radiation group (group R). The apoptosis rate and cell cycle were measured by flow cytometry. Results MTT assay showed that the inhibitory rate of CNE cells in nasopharyngeal carcinoma was positively correlated with the concentration of Apo-2L, and that the apoptosis rate of CNE cells in 100ng/ml group was significantly higher than that in other groups (P 0.05), and the ratio of G 2 / M phase cells in Apo-2L radiation group was significantly lower than that in other groups. Compared with other groups, there was a significant difference (P 0.05). Conclusion Apo-2L can inhibit proliferation and promote apoptosis of CNE cells cultured in vitro. The concentration of Apo-2L in combination with radiation can significantly increase the apoptosis rate of CNE cells and promote the synchronous distribution of cell cycle.
