【摘要】：目的研究色素播散綜合征(PDS)和色素性青光眼(PG)致病基因糖蛋白神經(jīng)調(diào)節(jié)肽B相關(guān)基因(Gpnmb)在BXD重組近交系(BXD RI)小鼠眼組織的表達(dá)差異及其基因調(diào)控網(wǎng)絡(luò)。方法采用Affymetrix基因芯片檢測(cè)確定68個(gè)品系的BXD RI小鼠眼組織中Gpnmb基因的表達(dá)差異。采用基因表達(dá)數(shù)量性狀基因座(eQTL)分析方法,結(jié)合GeneNetwork網(wǎng)站上的在線分析工具,研究差異表達(dá)的特征,進(jìn)而構(gòu)建Gpnmb基因調(diào)控網(wǎng)絡(luò)。結(jié)果 Gpnmb在BXD RI眼組織中平均表達(dá)水平為10.92,不同品系小鼠表達(dá)量存在差異。Gpnmb基因?yàn)轫樖阶饔玫膃QTL,End3基因是其下游候選基因�；诤虶pnmb高度相關(guān)基因的基因網(wǎng)絡(luò)得以構(gòu)建。結(jié)論構(gòu)建Gpnmb基因調(diào)控網(wǎng)絡(luò),為PDS和PG發(fā)病機(jī)制的深入研究提供線索。
[Abstract]:Objective to study the differential expression of glycoprotein neuromodulatory peptide B related gene (Gpnmb) related genes (Gpnmb) in the BXD recombinant inbred line (BXD RI) mouse eye tissues and its gene regulation network. Methods the Gpnmb gene in the BXD RI mouse eye tissues of 68 strains was determined by Affymetrix gene chip. The analysis method of gene expression quantitative trait loci (eQTL) and the online analysis tool on the GeneNetwork website were used to study the characteristics of the differential expression, and then the Gpnmb gene regulation network was constructed. The average expression level of Gpnmb in the BXD RI eye tissues was 10.92, and the expression of different strain mice was different in the.Gpnmb gene. For the CIS action of eQTL, the End3 gene is the downstream candidate gene. The gene network based on the highly related gene of Gpnmb is constructed. Conclusion the construction of the Gpnmb gene regulation network provides a clue for the in-depth study of the pathogenesis of PDS and PG.
