原發(fā)性開角型青光眼小梁網(wǎng)組織早衰的初步研究

【摘要】:【目的】觀察正常小梁網(wǎng)組織和POAG小梁網(wǎng)組織在超微結構和衰老相關β-半乳糖苷酶(SA-β-Gal)表達方面是否存在差異!痉椒ā空H诵×壕W(wǎng)組織取自廣東省眼庫的正常人眼球(10例),平均年齡(36.90±10.10)歲;POAG小梁網(wǎng)組織來自中山眼科中心青光眼科行小梁切除術者(10例),平均年齡(39.80±11.64)歲。用透射電鏡觀察兩組小梁網(wǎng)組織超微結構;用衰老相關β-半乳糖苷酶(SA-β-Gal)染色法對兩組小梁網(wǎng)組織進行染色,并分析兩者之間衰老染色的差異!窘Y果】透射電鏡發(fā)現(xiàn)正常人小梁網(wǎng)組織細胞膜完整,核染色均勻居中,胞漿中可見形態(tài)正常的中心體,周圍散落分布著的線粒體、溶酶體等細胞器;基底膜完整連續(xù);彈性纖維周圍繞有薄鞘,膠原纖維呈束狀排列、整齊;Schlemm管管腔通暢。而POAG患者的小梁網(wǎng)組織中可見細胞脫落溶解,細胞膜不完整,細胞核固縮或腫脹,線粒體水腫,線粒體嵴擴張;Schlemm管管腔變狹窄;彈性纖維增多且排列紊亂;基底膜增厚。SA-β-Gal染色發(fā)現(xiàn)POAG小梁網(wǎng)中的衰老細胞陽性率較正常人小梁網(wǎng)組織高(P0.001)。【結論】POAG小梁網(wǎng)超微結構破壞明顯,SA-β-Gal染色增強,在POAG小梁網(wǎng)中可能存在早衰。
[Abstract]:[objective] to observe the difference between normal trabecular meshwork and POAG trabecular reticulum in ultrastructure and senescence related 尾 -galactosidase SA- 尾 -Gal.Methods the normal trabecular meshwork was taken from the eye bank of Guangdong Province. There were 10 cases (mean age: 36.90 鹵10.10) years old (mean age: 36.90 鹵10.10) of the trabeculectomy in 10 patients with Ophthalmology. The average age was 39.80 鹵11.64 years old in 10 patients with trabeculectomy in Qingguang Eye Center of Zhongshan Ophthalmology Center. The ultrastructure of trabecular meshwork in two groups was observed by transmission electron microscope, and the tissue of trabecular meshwork was stained with senescence associated 尾 -galactosidase SA- 尾 -Gal) method. The difference of aging staining between them was analyzed. [results] Transmission electron microscope showed that the cell membrane of normal human trabecular meshwork was intact, the nucleus staining was uniform and the centrosome was seen in the cytoplasm, and the mitochondria were scattered around the cytoplasm. Lysosomes and other organelles; the basement membrane is complete and continuous; the elastic fibers are surrounded by thin sheath, the collagen fibers are arranged in bundles, and the Schlemm tube lumen is unobstructed. However, in the trabecular meshwork of POAG patients, the cells were exfoliated and dissolved, the cell membrane was incomplete, the nucleus was pyknosis or swelling, the mitochondria edema, the mitochondrial ridge dilated and the Schlemm tube lumen became narrow, the elastic fibers were increased and disordered. The positive rate of senescent cells in POAG trabecular meshwork was higher than that in normal human trabecular meshwork tissue by SA- 尾 -Gal staining. [conclusion] the ultrastructural damage of POAG trabecular meshwork was obviously enhanced by SA- 尾 -Gal staining, and there might be premature senescence in POAG trabecular meshwork.
