KAI1和COX-2在喉鱗癌組織中的表達(dá)及相關(guān)性研究

【摘要】:目的了解KAI1和COX-2在喉鱗癌組織中的表達(dá)的情況,探討二者在喉鱗癌中表達(dá)的意義及相關(guān)性,同時(shí)探討二者在喉鱗狀細(xì)胞癌發(fā)生發(fā)展及浸潤(rùn)轉(zhuǎn)移過(guò)程中的作用。方法應(yīng)用免疫組織化學(xué)SP法分別檢測(cè)45例經(jīng)病理證實(shí)喉鱗狀細(xì)胞癌患者的咽喉黏膜組織、10例正常體檢者咽喉黏膜組織中KAI1和COX-2的表達(dá)水平,分析二者表達(dá)水平及強(qiáng)度與病例的相關(guān)臨床指標(biāo)之間的關(guān)系,并分析這兩個(gè)基因之間在喉鱗狀細(xì)胞癌中表達(dá)的相關(guān)性。結(jié)果KAI1基因在喉鱗癌細(xì)胞中的表達(dá)低于在正常咽喉黏膜組織中的表達(dá)(χ~2=4.11, P 0.05);在喉癌組織中表達(dá)的陽(yáng)性率與年齡、性別、分化程度、臨床分期及是否有淋巴結(jié)轉(zhuǎn)移均無(wú)關(guān)(P0.05),但其陽(yáng)性表達(dá)強(qiáng)度與臨床分期及是否有淋巴結(jié)轉(zhuǎn)移有顯著相關(guān)性(P 0.05)。COX-2基因在喉鱗癌組織中高表達(dá),在正常咽喉黏膜組織中低表達(dá),差異有顯著性(P 0.01),在喉鱗癌組織中表達(dá)的陽(yáng)性率與性別、年齡及分化程度無(wú)關(guān),而與臨床分期及是否有淋巴結(jié)轉(zhuǎn)移有顯著相關(guān)性(P 0.05)。KAI1和COX-2在喉鱗癌組織中的表達(dá)呈負(fù)相關(guān)(關(guān)聯(lián)系數(shù)r=-0.30,P 0.05)。結(jié)論KAI1的表達(dá)在喉鱗癌的發(fā)生及浸潤(rùn)轉(zhuǎn)移過(guò)程中減弱或丟失,說(shuō)明其在喉鱗癌的發(fā)生、發(fā)展過(guò)程中具有一定抑制作用,可作為今后預(yù)測(cè)腫瘤細(xì)胞轉(zhuǎn)移潛能的指標(biāo);COX-2在喉鱗癌的發(fā)生、發(fā)展過(guò)程中表達(dá)增強(qiáng),,同樣說(shuō)明其在喉鱗癌細(xì)胞的發(fā)生發(fā)展及浸潤(rùn)轉(zhuǎn)移過(guò)程中起一定的促進(jìn)作用,也可作為今后檢測(cè)腫瘤細(xì)胞發(fā)生和轉(zhuǎn)移潛能的指標(biāo)之一。
[Abstract]:Objective to investigate the expression of KAI1 and COX-2 in laryngeal squamous cell carcinoma (LSCC) and to explore their significance and correlation in laryngeal squamous cell carcinoma (LSCC). Methods Immunohistochemical SP method was used to detect the expression of KAI1 and COX-2 in laryngeal mucosa of 45 patients with pathologically proved laryngeal squamous cell carcinoma (LSCC). To analyze the relationship between the expression level and intensity of the two genes and the clinical parameters of the patients, and to analyze the correlation between the expression of these two genes in laryngeal squamous cell carcinoma (LSCC). Results the expression of KAI1 gene in laryngeal squamous cell carcinoma cells was lower than that in normal laryngeal mucosa (蠂 ~ (2 +) 24.11, P 0.05), and the positive rate and age, sex, differentiation degree of KAI1 gene in laryngeal carcinoma tissue were significantly lower than those in normal laryngeal mucosa (蠂 ~ (2 +) 24.11, P 0.05). There was no correlation between clinical stage and lymph node metastasis, but there was a significant correlation between the positive expression of P 0.05).COX-2 gene and clinical stage and lymph node metastasis, and the low expression of P 0.05).COX-2 gene in normal pharynx and laryngeal mucosa. The positive rate of expression in laryngeal squamous cell carcinoma was not related to sex, age and degree of differentiation. The expression of P 0.05).KAI1 and COX-2 in laryngeal squamous cell carcinoma was negatively correlated with clinical stage and lymph node metastasis. Conclusion the expression of KAI1 is decreased or lost during the occurrence and invasion and metastasis of laryngeal squamous cell carcinoma, which indicates that it has a certain inhibitory effect on the occurrence and development of laryngeal squamous cell carcinoma. The expression of COX-2 in laryngeal squamous cell carcinoma (LSCC) may be used as a predictor of metastatic potential of tumor cells in the future, which also indicates that COX-2 plays an important role in the development, invasion and metastasis of laryngeal squamous cell carcinoma (LSCC). It can also be used as one of the indicators of tumor cell development and metastasis potential in the future.
