氟伐他汀對(duì)大鼠角膜堿燒傷后新生血管形成的影響

【摘要】:目的:探討氟伐他汀對(duì)大鼠角膜堿燒傷后新生血管形成的作用及其機(jī)制。方法:30只SD大鼠隨機(jī)分為3組:生理鹽水組、氟伐他汀組和地塞米松組,每組10只,均用1mol/L的NaOH燒傷大鼠右眼,生理鹽水組、氟伐他汀組和地塞米松組分別以生理鹽水、10-5 mol/L氟伐他汀和1g/L地塞米松點(diǎn)眼。堿燒傷后1-14d用裂隙燈觀察角膜的混濁度和新生血管的生長(zhǎng)情況,HE染色觀察角膜組織病理學(xué)改變,免疫組化觀察VEGF和CD31的表達(dá)情況。結(jié)果:堿燒傷后第3,7,14天,氟伐他汀組和地塞米松組的角膜混濁度評(píng)分和新生血管生長(zhǎng)評(píng)分都顯著低于生理鹽水組,差異有統(tǒng)計(jì)學(xué)意義,P0.05。組織病理學(xué)檢查結(jié)果提示,與生理鹽水組相比,氟伐他汀組和地塞米松組的角膜組織結(jié)構(gòu)明顯整齊,新生血管的形成明顯減少,伴少量炎性細(xì)胞浸潤(rùn)。免疫組化結(jié)果顯示氟伐他汀組和地塞米松組的VEGF和CD31的表達(dá)顯著減少。結(jié)論:氟伐他汀能減輕大鼠角膜堿燒傷,其機(jī)制可能是下調(diào)VEGF和CD31的表達(dá)。
[Abstract]:Aim: to investigate the effect and mechanism of fluvastatin on neovascularization after alkali burn of cornea in rats. Methods Thirty Sprague-Dawley rats were randomly divided into three groups: normal saline group, fluvastatin group and dexamethasone group with 10 rats in each group. All rats were treated with 1mol/L in the right eye and normal saline group. The eyes of fluvastatin group and dexamethasone group were treated with normal saline for 10-5 mol/L fluvastatin and 1g/L dexamethasone respectively. The corneal turbidities and neovascularization were observed by slit lamp from 1 to 14 days after alkali burn. The histopathological changes of cornea were observed by HE staining and the expression of VEGF and CD31 were observed by immunohistochemistry. Results: the corneal turbidity score and neovascularization score in fluvastatin group and dexamethasone group were significantly lower than those in saline group (P 0.05). Histopathological examination showed that the corneal tissue structure of fluvastatin group and dexamethasone group was obviously neat compared with normal saline group. The formation of neovascularization was obviously decreased and a small amount of inflammatory cells infiltrated in fluvastatin group and dexamethasone group. Immunohistochemical results showed that the expression of VEGF and CD31 in fluvastatin group and dexamethasone group decreased significantly. Conclusion: fluvastatin can attenuate the alkali burn of cornea in rats by down-regulating the expression of VEGF and CD31.
