脫細(xì)胞真皮基質(zhì)在角膜中植入性的生物力學(xué)研究

【摘要】:對(duì)脫細(xì)胞真皮基質(zhì)(acellular dermal matrix,ADM)囊袋植入兔角膜后的生物相容性、生物學(xué)轉(zhuǎn)歸及組織結(jié)構(gòu)改變進(jìn)行生物力學(xué)研究,以探討其作為組織工程角膜支架材料的可行性.對(duì)ADM進(jìn)行了系統(tǒng)的組織微觀結(jié)構(gòu)觀察和植入性實(shí)驗(yàn)研究,組織結(jié)構(gòu)觀察包括HE染色、膠原染色、免疫熒光染色和透射電鏡觀察.在植入實(shí)驗(yàn)中,將ADM按不同預(yù)處理方法分為無(wú)干預(yù)組、脫水組和復(fù)水組,將3組ADM囊袋植入兔角膜,觀察其術(shù)后不同時(shí)期在體和離體的生物相容性和組織學(xué)特性.結(jié)果發(fā)現(xiàn):ADM由橫縱交替、排列疏松的Ⅰ和Ⅲ型膠原纖維束構(gòu)成,未見(jiàn)細(xì)胞結(jié)構(gòu);囊袋植入角膜后有不同程度的炎癥反應(yīng),部分植片透明;組織學(xué)觀察顯示3組ADM植入角膜后均有細(xì)胞長(zhǎng)入,并進(jìn)行了膠原纖維重塑.研究工作表明,經(jīng)適當(dāng)預(yù)處理且結(jié)構(gòu)致密的脫細(xì)胞真皮基質(zhì)植入后可以長(zhǎng)期穩(wěn)定存留于角膜內(nèi),是一種比較理想的組織工程角膜支架.
[Abstract]:The biocompatibility, biological outcome and changes of tissue structure of acellular dermal matrixadm in rabbit cornea were studied by biomechanical study in order to explore its feasibility as a tissue engineering corneal scaffold.The microstructures and implants of ADM were observed systematically, including HE staining, collagen staining, immunofluorescence staining and transmission electron microscopy.In the implantation experiment, ADM was divided into three groups according to different pretreatment methods: no intervention group, dehydration group and rehydration group. The three groups of ADM bag were implanted into the cornea of rabbits. The biocompatibility and histological characteristics of ADM were observed in vitro and in vitro at different time after operation.The results showed that: ADM was composed of loose collagen bundles of type 鈪,
