細(xì)胞色素C在老年SD大鼠耳蝸不同階段凋亡壞死毛細(xì)胞中表達(dá)的變化

【摘要】:目的:觀察細(xì)胞色素C在老年SD大鼠不同階段凋亡、壞死毛細(xì)胞中表達(dá)的變化,探討老年大鼠耳蝸毛細(xì)胞死亡的機(jī)制。方法:16只SD大鼠分組:青年組8只,2~3月齡;老年組8只,24~26月齡。應(yīng)用電位反應(yīng)測聽儀檢測不同頻率短純音(4、8、16、32和40kHz)誘發(fā)的青年組和老年組大鼠雙側(cè)ABR。聽覺功能測試后,解剖取出雙耳聽泡,分離耳蝸基膜。分別用細(xì)胞核DNA染料碘化丙錠染色不同月齡大鼠耳蝸基膜細(xì)胞核,細(xì)胞色素C免疫熒光染料標(biāo)記大鼠耳蝸基膜細(xì)胞的細(xì)胞色素C蛋白,熒光顯微鏡下觀察不同月齡大鼠耳蝸基膜細(xì)胞熒光染色的變化。結(jié)果:老年組大鼠的不同頻率ABR閾值均高于青年組(P0.01)。微弱的細(xì)胞色素C熒光標(biāo)記物存在于青年組大鼠耳蝸基膜毛細(xì)胞質(zhì)(細(xì)胞膜的內(nèi)側(cè),線粒體存在部位),增強(qiáng)的細(xì)胞色素C免疫反應(yīng)物存在于老年組大鼠耳蝸基膜細(xì)胞。細(xì)胞色素C陽性反應(yīng)見于:①細(xì)胞核型不變;②碘化丙錠染色略有加深,細(xì)胞核型不變;③細(xì)胞核輕微核固縮;④核腫脹的毛細(xì)胞。相反,部分核固縮≤3/4核的毛細(xì)胞細(xì)胞色素C陽性反應(yīng)物減少或消失,核缺失區(qū)域的毛細(xì)胞未見細(xì)胞色素C陽性反應(yīng)物。老年大鼠耳蝸基膜凋亡毛細(xì)胞線粒體釋放的細(xì)胞色素C存在核內(nèi)轉(zhuǎn)移現(xiàn)象。結(jié)論:細(xì)胞色素C免疫熒光染色為老年耳蝸基膜毛細(xì)胞退行性變?cè)缙诘募?xì)胞生物學(xué)標(biāo)記之一。細(xì)胞色素C向細(xì)胞核內(nèi)的轉(zhuǎn)移,可能與老年大鼠耳蝸凋亡毛細(xì)胞染色質(zhì)凝聚相關(guān)。
[Abstract]:Objective: to observe the changes of cytochrome C in the apoptosis and necrotic hair cells of aged SD rats and to explore the mechanism of cochlear hair cell death in aged rats. Methods: sixteen SD rats were divided into two groups: young rats (n = 8) and young rats (n = 8) aged 23 months. The auditory function of the young rats and the aged rats of the young group and the aged group were measured by potentiometric audiometry. The auditory vesicles of both ears were taken out after the auditory function test. The basal membrane of cochlea was isolated and stained with nuclear DNA dye iodized propyl ingot. Cytochrome C immunofluorescence dye was used to label the cytochrome C protein in rat cochlear basement membrane cells. Fluorescence microscope was used to observe the changes of fluorescence staining of cochlear basement membrane cells in rats of different months. Results: the ABR thresholds of different frequencies in the aged rats were higher than those in the young group (P 0.01). The weak fluorescent labeled cytochrome C existed in the young group. The cytoplasm of the basal membrane of rat cochlea. The enhanced cytochrome C immunoreactive substance was found in the basal membrane cells of cochlea of the aged rats.
