納豆激酶對(duì)兔缺血型視網(wǎng)膜病變模型影響的實(shí)驗(yàn)研究

【摘要】:目的:用氬激光光凝視網(wǎng)膜中央靜脈制作兔視網(wǎng)膜中央靜脈阻塞(CRVO)模型,玻璃體腔注射納豆激酶評(píng)價(jià)其對(duì)視網(wǎng)膜的作用。 方法:采用氬激光直接光凝法造成兔CRVO模型,并通過眼底熒光素鈉血管造影(FFA)及眼底照相證實(shí)造模成功后將其分為3組,每組12只眼,納豆激酶A組連續(xù)3天玻璃體腔注射0.1 FU納豆激酶提取液0.1 ml;林格氏液對(duì)照B組同方法注射林格氏液0.1 ml;空白模型對(duì)照C組未外加處理。3組均在光凝前,光凝后進(jìn)行眼底照相,FFA檢測(cè);光凝前及光凝后2周3組兔進(jìn)行閃光視網(wǎng)膜電流圖(flash electroretinogram ,FERG)檢查和眼球壁病理切片蘇木精-伊紅染色觀察。 結(jié)果:①激光可以建立兔CRVO模型,并模擬了人的CRVO疾病發(fā)展進(jìn)程;造模成功后FFA顯示視網(wǎng)膜中央靜脈血流中斷,末梢血管充盈擴(kuò)張,②激光后2周,C組兔眼FERG a、b波潛伏期較造模前明顯延長,振幅降低;③激光后2周,A組FERG a、FERG b波潛伏期較B、C組明顯短,振幅較B、C組明顯大(P0.05)④與激光造模前相比,A組兔眼FERG a、FERG b波潛伏期延長,振幅降低;⑤A組與B組相比,病理切片觀察顯示前者視網(wǎng)膜細(xì)胞排列緊密,數(shù)量較多,后者有明顯網(wǎng)膜結(jié)構(gòu)疏松。 結(jié)論:氬激光致兔CRVO模型是可行成功的,FERG和病理結(jié)果提示納豆激酶可部分改善視網(wǎng)膜功能。
[Abstract]:Aim: to establish a rabbit model of central retinal vein occlusion (CRVOO) by argon laser photocoagulation, and to evaluate the role of nattokinase by intravitreal injection. Methods: the rabbit CRVO model was established by direct photocoagulation with argon laser. The model was confirmed by fundus fluorescein angiography (FFA) and fundus photography. The model was divided into 3 groups with 12 eyes in each group. In group A, 0.1 FU natto kinase extract was injected into vitreous cavity for 3 consecutive days; Linger's solution was injected with 0.1 ml in group B in the same way; group C in blank model group was treated with untreated 0.1 ml before photocoagulation. The fundus radiography (FFA) was performed before photocoagulation and 2 weeks after photocoagulation, and flash electroretinogram FERG and hematoxylin-eosin staining were performed in 3 groups of rabbits before and 2 weeks after photocoagulation. Results the CRVO model of rabbit could be established by 1: 1 laser, and the development process of human CRVO disease was simulated. After the successful establishment of the model, FFA showed the interruption of blood flow in the central retinal vein. In group C, the latency of FERG a b wave in group C was significantly longer than that in group C, and the latency of wave B in group A was shorter than that in group C 2 weeks after laser filling and dilatation of peripheral blood vessels, and the amplitudes decreased 2 weeks after laser irradiation, and the latency of wave B in group A was significantly shorter than that in group C. The amplitude of group A was significantly longer than that of group B (P 0.05) compared with that of group A (before laser modeling), and the latency of wave B in group A was longer than that of group A, and the amplitude of group A was lower than that of group B. The histopathological observation showed that the retinal cells of group A were arranged closely and the number of retinal cells was more than that of group B. The latter has a loose reticular membrane structure. Conclusion: rabbit CRVO model induced by argon laser is feasible and successful, and the pathological results suggest that nattokinase can partially improve retinal function.
