翼狀胬肉中血管內(nèi)皮生長因子、內(nèi)皮抑素的表達以及新生血管生成情況

【摘要】:目的通過研究翼狀胬肉組織中新生血管數(shù)量、促進血管生成因子以及抑制血管生長因子的表達情況,探討翼狀胬肉組織中促進血管形成和抑制血管生成的情況。方法手術獲得正常的結(jié)膜組織35例(對照組)及翼狀胬肉組織40例(實驗組),用免疫組化及Western blot的方法分別檢測兩組樣本中新生血管密度(MVD)、血管內(nèi)皮生長因子(VEGF)、內(nèi)皮抑素(Endostatin)的表達情況。其中,CD31是標記組織MVD的指標。結(jié)果①免疫組化法結(jié)果顯示:在翼狀胬肉組織中MVD的含量明顯高于正常結(jié)膜組織;VEGF在正常球結(jié)膜組織中僅有少量陽性表達,在翼狀胬肉組織中陽性細胞明顯增多,著色顯著加深;翼狀胬肉組織中上皮細胞基底細胞層中的Endostatin抗原陽性表達則明顯少于正常球結(jié)膜組織。②Western blot結(jié)果顯示人正常球結(jié)膜與翼狀胬肉中VEGF及Endostatin蛋白表達存在差異。翼狀胬肉組織中VEGF蛋白/β-actin(1.46±0.59)顯著高于正常球結(jié)膜組/β-actin(0.46±0.45),差異有統(tǒng)計學意義(P0.05);翼狀胬肉組織中Endostatin蛋白/β-actin(0.58±0.11)顯著低于正常球結(jié)膜組/β-actin(1.54±0.12),差異有統(tǒng)計學意義(P0.05)。結(jié)論在翼狀胬肉中發(fā)現(xiàn)其MVD及促血管生長因子的表達高于正常結(jié)膜組織,而血管抑制因子的表達含量降低,這一血管平衡的打破可能是翼狀胬肉的發(fā)病基礎之一。
[Abstract]:Objective to study the expression of angiogenic factors and angiogenesis factors in pterygium. Methods 35 cases of normal conjunctival tissue (control group) and 40 cases of pterygium tissue (experimental group) were obtained by immunohistochemistry and Western blot. Methods the expression of neovascularization density (MVD), vascular endothelial growth factor (VEGF) and endostatin (endostatin) in the two groups were detected respectively. The expression of MVD in normal conjunctiva was significantly higher than that in normal conjunctiva. In pterygium, the positive cells increased significantly and the staining deepened significantly. The positive expression of Endostatin antigen in the basal cell layer of epithelial cells in pterygium was significantly lower than that in the normal bulbar conjunctiva tissue. 2 Western blot results showed that there was a difference in the expression of VEGF and Endostatin protein between human normal bulbar conjunctiva and pterygium and pterygium pterygium. VEGF protein / 尾 -actinine in pterygium was significantly higher than that in conjunctiva conjunctiva / 尾 -actinine 0.46 鹵0.45, and Endostatin protein / 尾 -actinin 0.58 鹵0.11 in pterygium was significantly lower than that in normal conjunctiva / 尾 -actinine 1.54 鹵0.120.Conclusion it is found in pterygium that the protein / 尾 -actinin in pterygium is significantly lower than that in normal conjunctiva / 尾 -actinin (1.54 鹵0.12). Conclusion there is a significant difference in pterygium between pterygium and pterygium. The expression of MVD and vascular growth factor was higher than that of normal conjunctiva. The decrease of the expression of angiostatin may be one of the pathogenesis of pterygium.
