電針耳穴對(duì)豚鼠老年性聾耳蝸核、下丘和聽皮層中SOD表達(dá)的影響

【摘要】:目的:構(gòu)建D-半乳糖老年性聾豚鼠模型,探討超氧化物歧化酶(superoxide dismutase,SOD)在豚鼠老年性聾發(fā)病中的作用及電針耳穴對(duì)老年性聾的防治作用和機(jī)制。 方法:采用成年雜色豚鼠(體重300~550g)30只,隨機(jī)分成三組:對(duì)照組10只,D-半乳糖模型組10只,D-半乳糖+電針組10只。采用2年雜色豚鼠(體重600~900g)10只作為老年組。豚鼠飼養(yǎng)一周后,D-半乳糖模型組給予D-半乳糖(300mg/kg)頸背部皮下注射,每日一次,連續(xù)六周;D-半乳糖+電針組給予等比例體重的D-半乳糖頸背部皮下注射,同時(shí)給予電針聽宮和翳風(fēng)兩穴位,每日一次,連續(xù)六周;對(duì)照組給予等量生理鹽水頸背部皮下注射,每日一次,連續(xù)六周;老年組低噪聲環(huán)境下飼養(yǎng)。造模制備后于我科門診聽力學(xué)中心進(jìn)行聽性腦干反應(yīng)(ABR)測定后處死,,取每組豚鼠的耳蝸核、下丘和聽皮層,考馬斯亮蘭測定蛋白含量,分光光度儀檢測上述組織中SOD的表達(dá)情況,計(jì)算SOD的活性。各組部分聽皮層迅速置于4%多聚甲醛液中固定,石蠟切片HE染色觀察各組中聽皮層神經(jīng)元的形態(tài)。數(shù)據(jù)采用方差分析方法進(jìn)行統(tǒng)計(jì)學(xué)分析。 結(jié)果:1. D-半乳糖模型組和老年組豚鼠均出現(xiàn)老化癥狀:聽力逐漸減退、食欲下降和脫毛等現(xiàn)象提示造模成功。2. ABR測定結(jié)果:正常對(duì)照組的ABR閾值為39.00±7.38dB SPL. D-半乳糖模型組、D-半乳糖+電針組和老年組的ABR閾值分別79.44±6.62、70.00±5.88和81.88±4.43dB SPL(各實(shí)驗(yàn)組和老年組與正常對(duì)照組比較,P<0.001)。3.超氧化物歧化酶SOD活性測定:與正常對(duì)照組比較,D-半乳糖模型組和老年組,耳蝸核、下丘和聽皮層中SOD活性均下降,差異有統(tǒng)計(jì)學(xué)意義(P<0.01);與D-半乳糖模型組比較,D-半乳糖+電針組,三個(gè)部位的SOD活性均增高(P<0.01)。 結(jié)論:1. D-半乳糖300mg/kg頸背部皮下注射6周可建立老年性聾豚鼠模型。2. D-半乳糖可以導(dǎo)致豚鼠聽覺中樞的老化。3.聽覺中樞的老化可能與SOD活性降低所導(dǎo)致的抗氧化功能下降有關(guān)。4.針刺聽宮和翳風(fēng)兩個(gè)穴位可在一定程度上抑制D-半乳糖所致的豚鼠聽覺中樞的老化過程。
[Abstract]:Objective: to study the role of superoxide dismutase (SOD) in the pathogenesis of presbycusis in guinea pigs and the preventive effect and mechanism of electroacupuncture on presbycusis. Methods: 30 adult heterochromatic guinea pigs were used. Three groups were randomly divided into three groups: control group (n = 10) with D- galactose electroacupuncture (n = 10). Ten pigmented guinea pigs (body weight 600,900g) were used as aged group. After one week of feeding, Dgalactose was given to Dgalactose model group (D- galactose group). Sugar is 300 mg / kg) subcutaneously injected into the neck and back, Once a day for six weeks, the rats in the Dgalactose electroacupuncture group were given subcutaneous injection of D- galactose at the neck and back of the same proportion of body weight, at the same time, they were given electroacupuncture at auricula and Yifeng, once a day for six weeks. The control group was given subcutaneous injection of normal saline once a day for six weeks, and the aged group was fed in low noise environment. The model was made and measured at the auditory brainstem response (ABR) of our outpatient department. The cochlear nucleus, inferior colliculus and auditory cortex of each group were taken. Coomassie brilliant blue was used to determine the protein content, the expression of SOD in the tissues was detected by spectrophotometry, and the activity of SOD was calculated. Part of the auditory cortex in each group was quickly fixed in 4% paraformaldehyde solution. The morphology of auditory cortex neurons in each group was observed by HE staining in paraffin sections, and the data were analyzed statistically by ANOVA. Results the aging symptoms were observed in both the D-galactose model group and the senile group. (2) the results of ABR determination: the ABR threshold of normal control group is 39.00 鹵7.38dB SPL.D- galactose electroacupuncture group and senile group have ABR threshold of 79.44 鹵6.62% 70.00 鹵5.88 and 81.88 鹵4.43 dB respectively. The activity of superoxide dismutase (SOD) was measured in the old group and the model group of D-galactose compared with the normal control group (P < 0.001), and the activity of superoxide dismutase (SOD) in the aged group was higher than that in the control group. The activity of SOD in the cochlear nucleus, inferior colliculus and auditory cortex decreased significantly (P < 0.01), and the SOD activity in the three parts of the group was significantly higher than that in the Dgalactose model group (P < 0.01), while that in the Dgalactose electroacupuncture group was significantly higher than that in the Dgalactose model group (P < 0.01). ConclusionDgalactose 300mg / kg subcutaneously injected into the neck and back for 6 weeks can establish the guinea pig model of presbycusis. 2.Dgalactose can lead to the aging of auditory center of guinea pig. 3. The aging of auditory center and the decrease of SOD activity may lead to the antioxidation of guinea pig auditory center. Acupuncture of Acupoint and Yifeng can inhibit the aging process of hearing center induced by D-galactose in guinea pigs to some extent.
