MK-801拮抗水楊酸鈉致螺旋神經(jīng)節(jié)細(xì)胞損傷的實(shí)驗(yàn)研究

【摘要】:目的探討非特異性阻斷NMDA受體拮抗水楊酸鈉對(duì)離體培養(yǎng)螺旋神經(jīng)節(jié)細(xì)胞興奮性損傷的作用。方法將培養(yǎng)的耳蝸螺旋神經(jīng)節(jié)細(xì)胞隨機(jī)分為3組,分別為正常對(duì)照組:培養(yǎng)液中僅加入1mM谷氨酸;水楊酸鈉組:1mM谷氨酸+5mM水楊酸鈉;MK-801組:50μM MK-801+1mM谷氨酸+5mM水楊酸鈉。培養(yǎng)24h后加藥物干預(yù)3h,收集細(xì)胞采用實(shí)時(shí)熒光定量PCR及免疫熒光技術(shù)檢測(cè)BDNF exonⅣ,BDNF exonⅥ轉(zhuǎn)錄及Caspase-3mRNA轉(zhuǎn)錄和蛋白表達(dá)的改變,研究應(yīng)用MK-801非特異性阻斷NMDA受體拮抗水楊酸鈉對(duì)離體培養(yǎng)螺旋神經(jīng)節(jié)細(xì)胞興奮性損傷的作用。結(jié)果水楊酸鈉組較谷氨酸組及MK-801組螺旋神經(jīng)節(jié)細(xì)胞中BDNF exonⅣ,BDNF exonⅥ和Caspase-3 mRNA的轉(zhuǎn)錄及Cas-pase-3蛋白的表達(dá)水平顯著增高(P值均0.05);BDNF exonⅣ的轉(zhuǎn)錄在MK-801組較谷氨酸組明顯降低(P0.05);BDNF exonⅥ的轉(zhuǎn)錄在MK-801組較谷氨酸組未發(fā)生具有統(tǒng)計(jì)學(xué)意義的改變;Caspase-3的轉(zhuǎn)錄及蛋白的表達(dá)在MK-801組較谷氨酸組顯著提高(P0.05)。結(jié)論非特異性阻斷NMDA受體能拮抗水楊酸鈉引起的離體培養(yǎng)螺旋神經(jīng)節(jié)細(xì)胞中BDNFexonⅣ,BDNFexonⅥ及Caspase-3上調(diào)。
[Abstract]:Objective to investigate the antagonistic effect of sodium salicylate on excitatory injury induced by sodium salicylate by blocking NMDA receptor in vitro. Methods cultured cochlear spiral ganglion cells were randomly divided into three groups.
