化學(xué)計量學(xué)方法在抗腫瘤活性成分篩選中的應(yīng)用

【摘要】: 粘細(xì)菌可產(chǎn)生豐富的次級代謝產(chǎn)物,其代謝產(chǎn)物的化學(xué)結(jié)構(gòu)和生物活性作用機(jī)制的多樣性可以與著名的鏈霉菌類群相媲美,埃博霉素是人們最為熟知的代謝產(chǎn)物之一,但這些代謝產(chǎn)物產(chǎn)量一般較低且產(chǎn)量不穩(wěn)定,進(jìn)行逐個分離純化和活性的測定非常困難;瘜W(xué)計量學(xué)是將統(tǒng)計學(xué)、數(shù)學(xué)和計算機(jī)科學(xué)綜合應(yīng)用在化學(xué)分析領(lǐng)域的新興學(xué)科,本文將這種方法用于纖維堆囊菌次級代謝中活性成分的初步篩選中。具體方法是收集不同發(fā)酵條件和利用不同發(fā)酵培養(yǎng)基的纖維堆囊菌發(fā)酵液,提取其次級代謝物粗提液樣品,分別測定樣品的抗人鼻咽癌腫瘤細(xì)胞CNE活性、抗舌磷癌細(xì)胞Tca-8113活性和抗人乳腺導(dǎo)管ZR-75-1活性,同時采用高效液相色譜法(HPLC)測定樣品中各組分,采用化學(xué)計量學(xué)方法中的多元線性回歸方法建立樣品中各組HPLC分峰面積分別與其抗人鼻咽癌腫瘤細(xì)胞CNE活性、抗舌磷癌細(xì)胞Tca-8113活性和抗人乳腺導(dǎo)管ZR-75-1活性間的相關(guān)模型,并采用t檢驗方法,檢驗各模型中各項系數(shù)的顯著性,以篩選顯著影響樣品抗人鼻咽癌腫瘤細(xì)胞CNE活性、抗舌磷癌細(xì)胞Tca-8113活性和抗人乳腺導(dǎo)管ZR-75-1活性的HPLC峰所對應(yīng)的組分,為纖維堆囊菌活性次級代謝產(chǎn)物的抗腫瘤活性成分的篩選提供了理論依據(jù)。
[Abstract]:Myxobacteria can produce abundant secondary metabolites, and their chemical structure and biological activity mechanism can be compared with those of the famous Streptomyces, and Ebomycin is one of the most well-known metabolites. However, the yield of these metabolites is generally low and the yield is unstable, so it is very difficult to separate, purify and determine the activity one by one. Chemometrics is an emerging discipline that integrates statistics, mathematics and computer science into the field of chemical analysis. In this paper, this method was applied to the screening of active components in the secondary metabolism of Cystis cellulosae. The specific method was to collect the fermentation broth with different fermentation conditions and different fermentation media, and to extract samples from the crude extracts of secondary metabolites. The CNE activity, Tca-8113 activity and ZR-75-1 activity of anti-human nasopharyngeal carcinoma (NPC) tumor cells, anti-tongue phosphorous cancer cells and human breast ducts were determined by HPLC, respectively, and each component of the samples was determined by high performance liquid chromatography (HPLC). The multivariate linear regression method in chemometrics was used to establish a model of correlation between peak area of HPLC and CNE activity, Tca-8113 activity and ZR-75-1 activity of human mammary duct in human nasopharyngeal carcinoma cell line. T test method was used to test the significance of the coefficients in each model, in order to screen the components corresponding to the HPLC peak, which significantly affected the CNE activity, the Tca-8113 activity and the ZR-75-1 activity of the tumor cells of human nasopharyngeal carcinoma (NPC), the anti-tongue phosphorous cancer cells and the anti-human mammary duct ZR-75-1. It provides a theoretical basis for the screening of anti-tumor active components of the secondary metabolites of cystobacterium fibrillaris.
