Eotaxin及CCR3在鼻息肉中的表達及其意義

【摘要】:目的:采用免疫組化染色方法觀察在人鼻息肉中嗜酸粒細(xì)胞趨化因子(Eotaxin)與嗜酸性粒細(xì)胞趨化因子受體(CCehemokine receptor3,CCR3)的表達情況,探討其臨床意義。 方法:選擇鼻息肉組織28例,鼻中隔矯正術(shù)患者的下鼻甲粘膜組織11例作為對照組,采用免疫組織化學(xué)染色方法(SABA法)觀察Eotaxin與CCR3在鼻息肉與對照組中的表達,并進行統(tǒng)計學(xué)分析 結(jié)果: 1.嗜酸性粒細(xì)胞(eosinophieis, EOS)浸潤數(shù):HE染色可見鼻息肉組織黏膜層、黏膜下層及血管和腺體周圍組織中可見各種炎性細(xì)胞浸潤,其中以EOS浸潤為主(EOS計數(shù):55.64±15.11)。對照組下鼻甲黏膜中可見少量的嗜酸性粒細(xì)胞浸潤(EOS計數(shù):5.27±3.72)。EOS在鼻息肉組中的浸潤數(shù)顯著高于對照組,差異有統(tǒng)計學(xué)意義(P0.01)。 2.免疫組織化學(xué)染色片上可見Eotaxin陽性表達位于細(xì)胞漿中,呈棕黃色,其分布廣,主要表達在黏膜上皮細(xì)胞、血管內(nèi)皮細(xì)胞、腺體細(xì)胞及各種炎性細(xì)胞(Eotaxin陽性細(xì)胞數(shù):69.28±19.48);在對照組中陽性染色弱,分布于黏膜上皮或浸潤的炎性細(xì)胞中(Eotaxin陽性細(xì)胞數(shù):3.82±2.40)。在鼻息肉組中的表達顯著多于對照組,且差異有統(tǒng)計學(xué)意義(P0.01)。 3.免疫組織化學(xué)染色片中細(xì)胞膜染成棕黃色的為CCR3陽性細(xì)胞,在鼻息肉組織中主要表達在黏膜上皮細(xì)胞與黏膜下固有層中浸潤的EOS細(xì)胞膜上并且染色較深(CCR3陽性表達數(shù):55.79±21.60)。在對照組下鼻甲黏膜中可見少量的CCR3陽性細(xì)胞并且染色淺(CCR3陽性表達數(shù):7.45±5.10)。鼻息肉組CCR3的陽性表達數(shù)顯著多于對照組,且差異有統(tǒng)計學(xué)意義(P0.01)。 4.Eotaxin陽性表達數(shù)與浸潤的EOS數(shù)量呈顯著正相關(guān)(r值為0.883;P值為0.00),具有統(tǒng)計學(xué)意義(P0.01)。CCR3陽性表達數(shù)和嗜酸粒細(xì)胞數(shù)呈顯著正相關(guān)(r值為0.800,P值為0.00),具有統(tǒng)計學(xué)意義(P0.01)。Eotaxin與CCR3陽性表達數(shù)呈顯著正相關(guān)(r值為0.776,P值為0.00),具有統(tǒng)計學(xué)意義(P0.01)。 結(jié)論:鼻息肉中可見大量的EOS浸潤,Eotaxin和CCR3的表達均明顯增強,Eotaxin和CCR3可能參與了EOS的遷移和聚集,并在鼻息肉形成過程中發(fā)揮重要作用。
[Abstract]:Objective: to investigate the expression of eotaxin and CCehemokine receptor 3 CCR3 in human nasal polyps by immunohistochemical staining. Methods: 28 cases of nasal polyps and 11 cases of nasal septum correction were selected as control group. The expression of Eotaxin and CCR3 in nasal polyps and control group was observed by immunohistochemical staining. And statistical analysis. Results:. 1. The infiltration of eosinophieis (EOS) in eosinophieis (eosinophieis) was observed in nasal polyp tissue mucosal layer, submucosal layer, blood vessel and periglandular tissue. The number of eosinophils infiltrated in the mucosa of the inferior turbinate in the control group was significantly higher than that in the control group (P 0.01), and the number of eosinophils infiltrated in the nasal polyp group was significantly higher than that in the control group (P < 0.01), and a small amount of eosinophil infiltration was found in the mucosa of the inferior turbinate in the control group. 2. The positive expression of Eotaxin was found in the cytoplasm of the cells, which was brown and widely distributed, mainly in the epithelial cells of the mucous membrane and the endothelial cells of the blood vessel. The number of eotaxin positive cells in glandular cells and various inflammatory cells was 69.28 鹵19.48, and the number of eotaxin positive cells in mucosal epithelium or infiltrated inflammatory cells was significantly higher in nasal polyp group than in control group, and the number of eotaxin positive cells in mucosal epithelium or infiltrated inflammatory cells was significantly higher than that in control group. The difference was statistically significant (P 0.01). 3. CCR3 positive cells were stained with brown cell membrane in immunohistochemical staining tablets. In nasal polyps, it was mainly expressed on EOS cell membrane infiltrated in mucosal epithelial cells and submucous lamina propria and stained deeply. The positive expression of CCR3 in nasal polyps was 55.79 鹵21.600.A small number of CCR3 positive cells were observed and stained in the mucosa of inferior turbinate in control group. The positive expression of CCR3 in the nasal polyp group was significantly higher than that in the control group, and the positive expression of CCR3 in the nasal polyp group was significantly higher than that in the control group. The difference was statistically significant (P 0.01). 4. There was a significant positive correlation between eotaxin positive expression and the number of infiltrated EOS. The positive correlation between the positive expression of eotaxin and the number of eosinophils was 0.800, P = 0.000.The positive correlation between eotaxin and the number of infiltrated EOS was 0.883 (P = 0.88) (P < 0.05), and the positive correlation between the positive expression of eotaxin and the number of eosinophilic granulocytes was 0.800 (P = 0.004). There was statistical significance between eotaxin and CCR3 positive. There was a significant positive correlation between the number of sexual expression and P value (P = 0.776P = 0.001), which was statistically significant (P 0.01). Conclusion: a large number of EOS infiltrating eotaxin and CCR3 expressions in nasal polyps may be involved in the migration and aggregation of EOS and play an important role in the formation of nasal polyps.
