Notch基因在變應(yīng)性鼻炎小鼠模型中的表達(dá)及其意義

【摘要】:目的:探討Notch信號(hào)通路相關(guān)基因在變應(yīng)性鼻炎模型小鼠鼻黏膜及外周血單個(gè)核細(xì)胞(PBMC)中的表達(dá)及意義。方法:7~8周齡BALB/c小鼠16只,隨機(jī)分成對(duì)照組和模型組。以卵清蛋白作為過敏原,建立變應(yīng)性鼻炎小鼠模型,分析比較小鼠過敏癥狀評(píng)分,采用蘇木精-伊紅染色分析比較鼻黏膜病理變化及炎性細(xì)胞的浸潤(rùn)程度,采用酶聯(lián)免疫吸附法檢測(cè)并分析小鼠外周血血清中IgE的表達(dá)水平。熒光定量聚合酶鏈反應(yīng)和流式細(xì)胞術(shù)從不同水平檢測(cè)模型組小鼠鼻黏膜及PBMC中Notch信號(hào)通路相關(guān)受體(Notch1~Notch4)的表達(dá)量,并與對(duì)照組比較。結(jié)果:成功建立了變應(yīng)性鼻炎BALB/c小鼠模型。模型組小鼠鼻黏膜中Notch1、Notch3、Notch4mRNA的表達(dá)明顯高于對(duì)照組(P0.01),而Notch2mRNA的表達(dá)卻低于對(duì)照組(P0.05);與此一致,模型組小鼠PBMC中Notch1、Notch3、Notch4的蛋白表達(dá)也明顯高于對(duì)照組(P0.01),而Notch2的蛋白卻低于對(duì)照組(P0.05)。結(jié)論:Notch基因的表達(dá)在變應(yīng)性鼻炎模型小鼠發(fā)病過程中有顯著改變,可能參與變應(yīng)性鼻炎的發(fā)生發(fā)展,為深入研究變應(yīng)性鼻炎的發(fā)病機(jī)制提供了思路。
[Abstract]:Objective: to investigate the expression and significance of Notch signaling pathway related genes in nasal mucosa and peripheral blood mononuclear cells (PBMC) of allergic rhinitis mice. The mouse model of allergic rhinitis was established with ovalbumin as allergen, and the allergic symptom score of mice was analyzed and compared. The pathological changes of nasal mucosa and the degree of infiltration of inflammatory cells were analyzed by hematoxylin and eosin staining. The expression of IgE in peripheral blood of mice was detected and analyzed by enzyme-linked immunosorbent assay (Elisa). Fluorescence quantitative Polymerase chain reaction (FQ-PCR) and flow Cytometry (FCM) were used to detect the Notch signaling pathway in the nasal mucosa and PBMC of the model group from different levels. The expression of Notch1 (Notch4), Results: the BALB/c mouse model of allergic rhinitis was successfully established. The expression of Notch1ntch3ntch4 mRNA in the nasal mucosa of mice in the model group was significantly higher than that in the control group (P 0.01), but the expression of Notch2mRNA was lower than that in the control group (P 0.05). The protein expression of Notch1, Notch3, Notch4 in PBMC of the model group was also significantly higher than that of the control group, while the protein of Notch2 was lower than that of the control group. Conclusion the expression of the 1 / Notch gene in allergic rhinitis model mice was significantly changed during the pathogenesis of allergic rhinitis. It may be involved in the occurrence and development of allergic rhinitis, which provides a way to further study the pathogenesis of allergic rhinitis.
