耳蝸核前腹核叢細(xì)胞的功能修飾先于聽覺的建立

【摘要】:目的通過全細(xì)胞膜片鉗技術(shù)研究新生SD乳鼠耳蝸核前腹核(AVCN)中叢細(xì)胞在發(fā)育過程中的電生理特性變化。方法運(yùn)用全細(xì)胞膜片鉗記錄出生后5~21 d的SD乳鼠耳蝸核前腹核中叢細(xì)胞的動(dòng)作電位(AP)反應(yīng)及微小興奮性突觸后電流(mEPSC),再通過相關(guān)數(shù)據(jù)采集及分析軟件分析動(dòng)作電位半峰寬和mEPSC的10%~90%上升時(shí)間(10%~90%Risetime)及時(shí)間常數(shù),并對(duì)此做進(jìn)一步統(tǒng)計(jì)分析處理,進(jìn)而比較不同年齡段新生SD乳鼠的耳蝸核前腹核中叢細(xì)胞的電生理反應(yīng)特性。結(jié)果在叢細(xì)胞動(dòng)作電位半峰寬的統(tǒng)計(jì)分析中發(fā)現(xiàn),叢細(xì)胞動(dòng)作電位的時(shí)間精確性隨著出生后年齡的增加而增加,在聽覺建立(出生后12 d)附近變化停止。叢細(xì)胞mEPSC的時(shí)間精確性也隨著出生后年齡的增加而增加,在聽覺建立附近變化停止。結(jié)論耳蝸核前腹核中叢細(xì)胞功能的修飾先于聽覺的建立。
[Abstract]:Objective to study the electrophysiological characteristics of plexus cells in the anterior ventral nucleus of the cochlear nucleus (AVCNN) of newborn SD rats by whole-cell patch clamp technique. Methods whole cell patch clamp was used to record the precochlear nucleus precochlear nucleus of neonatal SD rats at 5 to 21 days after birth. The action potential (APP) response and the minimal excitatory postsynaptic current (mEPSCN) of the plexus cells in the ventral nucleus were analyzed. The half peak width of the action potential and the 1090% rise time of mEPSC and the time constant were analyzed by the software of relevant data acquisition and analysis. The electrophysiological responses of the plexus cells in the anterior ventral nucleus of the cochlear nucleus of newborn SD rats of different ages were compared. Results in the statistical analysis of the half peak width of the action potential of the plexus cells, the electrophysiological response of the plexus cells in the anterior ventral nucleus of the cochlear nucleus was compared. The temporal accuracy of the action potential of plexus cells increased with the increase of postnatal age, and stopped in the vicinity of auditory establishment (12 days after birth). The temporal accuracy of mEPSC of plexus cells also increased with the increase of postnatal age. Conclusion the functional modification of the plexus cells in the anterior ventral nucleus of the cochlear nucleus is prior to the establishment of the auditory system.
