【摘要】：目的探討髓源性抑制細(xì)胞(MDSCs)在鼻咽癌患者外周血中的表達(dá)水平及其臨床意義,并在荷瘤小鼠體內(nèi)探討MDSCs可能的作用。方法采用流式細(xì)胞術(shù)分析49例鼻咽癌患者外周血及荷瘤小鼠脾臟MDSCs的比例,采用ELISA分析鼻咽癌患者及荷瘤小鼠中MDSCs對(duì)T細(xì)胞分泌細(xì)胞因子的影響。結(jié)果鼻咽癌患者外周血中MDSCs的比例顯著高于健康對(duì)照組,并且鼻咽癌患者外周血中MDSCs的比例與腫瘤的分期有關(guān),晚期患者外周血中MDSCs的比例高于早期患者;鼻咽癌荷瘤小鼠脾臟內(nèi)MDSCs的數(shù)量明顯高于對(duì)照組;研究表明MDSCs可明顯抑制T細(xì)胞分泌細(xì)胞因子。結(jié)論鼻咽癌患者外周血中MDSCs的比例較健康人高,且與鼻咽癌分期密切相關(guān),荷瘤小鼠實(shí)驗(yàn)表明腫瘤組小鼠脾臟MDSCs的數(shù)量明顯高于對(duì)照組,并且MDSCs可影響荷瘤小鼠細(xì)胞因子的分泌。MDSCs可能作為鼻咽癌生物治療的1個(gè)靶點(diǎn)。
[Abstract]:Objective to investigate the expression of myelogenous inhibitory cell (MDSCs) in peripheral blood of patients with nasopharyngeal carcinoma (NPC) and its clinical significance. Methods flow cytometry was used to analyze the proportion of MDSCs in peripheral blood and spleen of 49 patients with nasopharyngeal carcinoma. ELISA was used to analyze the effect of MDSCs on T cell cytokines secretion in nasopharyngeal carcinoma patients and tumor-bearing mice. Results the percentage of MDSCs in peripheral blood of NPC patients was significantly higher than that of healthy controls. The proportion of MDSCs in peripheral blood of patients with nasopharyngeal carcinoma was related to the stage of tumor, and the proportion of MDSCs in peripheral blood of patients with advanced stage was higher than that of patients with early stage. The amount of MDSCs in the spleen of nasopharyngeal carcinoma bearing mice was significantly higher than that of control group. Conclusion the proportion of MDSCs in peripheral blood of patients with nasopharyngeal carcinoma is higher than that of healthy people, and it is closely related to the stage of nasopharyngeal carcinoma. Tumor-bearing mice experiment showed that the number of spleen MDSCs in the tumor group was significantly higher than that in the control group. And MDSCs can affect cytokine secretion in tumor-bearing mice. MDSCs may be a target for nasopharyngeal carcinoma biotherapy.
