【摘要】：目的觀察形覺剝奪性弱視動(dòng)物模型應(yīng)用左旋多巴(levodopa,LD)治療后視皮質(zhì)17區(qū)生長相關(guān)蛋白-43(growth associated protein-43,GAP-43)的表達(dá),探討LD對(duì)形覺剝奪性弱視視覺功能恢復(fù)作用的可能機(jī)制。方法50只4周齡幼貓,隨機(jī)分為正常組、剝奪組、對(duì)照組、低劑量LD組及高劑量LD組共5組,每組10只。通過單眼眼瞼縫合制備形覺剝奪的弱視模型,觀察不同干預(yù)條件下各組圖形視覺誘發(fā)電位(pattern visual evoked potential,PVEP)的P100波的峰時(shí)及振幅的變化。應(yīng)用免疫組織化學(xué)法測定各組視皮層17區(qū)GAP-43的差異。結(jié)果8周齡時(shí),剝奪組、對(duì)照組、低劑量LD組、高劑量LD組剝奪眼P100波振幅比對(duì)側(cè)眼及同周齡正常組幅值降低,潛時(shí)延長(均為P0.05);12周齡時(shí),低劑量LD組剝奪眼P100波振幅增高,與對(duì)側(cè)眼及同周齡正常組相比差異均無統(tǒng)計(jì)學(xué)意義(均為P0.05),而P100波潛時(shí)則未達(dá)到正常(P0.05);高劑量LD組剝奪眼P100波振幅增高、潛時(shí)縮短,與對(duì)側(cè)眼及同周齡正常組相比差異均無統(tǒng)計(jì)學(xué)意義(均為P0.05)。8周齡時(shí)剝奪組視皮質(zhì)17區(qū)Ⅱ/Ⅲ、Ⅳ、Ⅴ/Ⅵ層GAP-43免疫陽性細(xì)胞密度均低于正常組,差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05);12周齡時(shí)高劑量LD組各層GAP-43免疫陽性細(xì)胞密度與正常組基本相等,差異無統(tǒng)計(jì)學(xué)意義(P0.05);對(duì)照組、低劑量LD組和正常組相比差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05)。結(jié)論GAP-43在形覺剝奪性弱視形成過程中起一定的作用,LD對(duì)形覺剝奪性弱視視功能改善的機(jī)制是通過GAP-43表達(dá)的改變而實(shí)現(xiàn)的。
