【摘要】：目的：研究兩種基質(zhì)金屬蛋白酶（MMP-1和MMP-2）在中耳膽脂瘤上皮組織中的表達與侵襲破壞的關系，并探討二者的相關性，從分子水平了解膽脂瘤的發(fā)病機制。方法：采用免疫組化法檢測39例經(jīng)病理切片證實為膽脂瘤型中耳炎患者不同部位的中耳膽脂瘤上皮標本中的MMP-1和MMP-2表達，，根據(jù)患者聽力學檢查分為輕度聾、中度聾、中重度聾、重度聾、極重度聾；根據(jù)術中所見將膽脂瘤侵襲情況分為Ⅰ度、Ⅱ度、Ⅲ度和Ⅳ度，結(jié)合患者臨床資料及膽脂瘤取材部位等進行統(tǒng)計分析。結(jié)果：不同的聽力損失等級、膽脂瘤侵襲情況的膽脂瘤上皮組織中MMP-1、MMP-2的陽性表達的差異有統(tǒng)計學意義（P＜0.05），MMP-1和MMP-2的表達呈正相關（r=0.831，P＜0.05）。性別、年齡，取材部位對MMP-1和MMP-2的表達均無統(tǒng)計學意義（P＞0.05）。結(jié)論：1．MMP-1和MMP-2在中耳膽脂瘤上皮中均有高表達，表明二者在膽脂瘤上皮的增殖及骨質(zhì)破壞的過程中都發(fā)揮重要作用。2．MMP-1和MMP-2在中耳膽脂瘤組織中隨著周圍骨質(zhì)破壞程度的范圍擴大和患者聽力損失程度的加重，二者表達量也隨之增高，提示二者在膽脂瘤上皮對骨質(zhì)的破壞行為和聽力損害中起非常重要的作用。3．MMP-1和MMP-2之間在中耳膽脂瘤上皮組織中的表達呈正相關，提示二者在膽脂瘤組織中的增殖和侵襲行為中可能有協(xié)同作用。
[Abstract]:Objective : To study the relationship between the expression of MMP - 1 and MMP - 2 in the epithelial tissue of middle ear cholesterin by two kinds of matrix metalloproteinases ( MMP - 1 and MMP - 2 ) . The expression of MMP - 1 and MMP - 2 was detected by immunohistochemistry . Results : The expression of MMP - 1 and MMP - 2 was positively correlated with the expression of MMP - 1 and MMP - 2 ( r = 0.831 , P < 0.05 ) . The expression of MMP - 1 and MMP - 2 was not statistically significant ( P > 0.05 ) . Conclusion : 1 . MMP - 1 and MMP - 2 were highly expressed in the epithelium of middle ear cholesterin , indicating that both MMP - 1 and MMP - 2 play an important role in the proliferation and destruction of tumor epithelial cells . The expression of MMP - 1 and MMP - 2 increased with the extent of peripheral bone destruction and the degree of hearing loss , suggesting that both of MMP - 1 and MMP - 2 play a very important role in the destruction of bone and hearing impairment . The expression of MMP - 1 and MMP - 2 was positively correlated with the expression of MMP - 1 and MMP - 2 in the epithelial tissue of the middle ear .
