【摘要】：目的：探討鼻息肉病人息肉組織中IL-5的表達(dá)情況,及其與Eos計(jì)數(shù)是否存在相關(guān)性。 方法：結(jié)合病史、鼻內(nèi)鏡、CT及化驗(yàn)室檢查將入選病人分為3組：對照組(13例),單純性鼻息肉組(22例),NP伴有過敏性鼻炎(allergic rhinitis, AR)組(20例)。正常對照組為單純鼻中隔偏曲患者,取部分肥大之中鼻甲。鼻內(nèi)鏡下取出息肉組織,切片保存。HE染色測定Eos計(jì)數(shù),免疫組化和酶聯(lián)免疫吸附法(enzyme-linked immuno sorbent assay, ELISA)檢測IL-5在組織中的表達(dá)情況。Eos計(jì)數(shù)和IL-5表達(dá)的組間差異采用單因素方差分析,Eos計(jì)數(shù)和IL-5表達(dá)的關(guān)聯(lián)性研究采用Pearson相關(guān)性分析。 結(jié)果： 1.HE染色：對照組鼻黏膜組織結(jié)構(gòu)清晰、完整,少見Eos浸潤；單純性NP組和NP伴AR組的組織明顯水腫,有大量的Eos浸潤。Eos計(jì)數(shù)分別為：對照組(1.2±0.4)/HP,單純性NP組(7.4±1.6)/HP,NP伴AR組(13.5±2.5)/HP。Eos計(jì)數(shù)具有組間差異性(F=180.1,P0.001)。 2.免疫組化染色：IL-5陽性染色細(xì)胞表現(xiàn)為胞漿呈棕黃色或棕褐色顆粒。對照組中未見明顯著色,單純性NP組中IL-5陽性染色細(xì)胞明顯可見,NP伴AR組可見大量的陽性染色細(xì)胞且著色加深(棕褐色)。對照組IL-5陽性染色細(xì)胞的平均灰度值為(177.3±21.1),單純性NP組為(124.8±7.4),NP伴AR組為(93.3±8.1),具有組間差異性(F=187.3,P0.001)。 3.IL-5濃度(ELISA法)：經(jīng)標(biāo)準(zhǔn)曲線計(jì)算出各組IL-5的濃度,分別為：對照組(1.3±0.2)n/ml,單純性NP組(2.9±0.3)ng/ml,NP伴AR組(4.8±0.5)ng/ml。IL-5濃度具有組間差異性(F=317.1,P0.001)。 4.IL-5表達(dá)與Eos計(jì)數(shù)的關(guān)系：對照組中Eos計(jì)數(shù)和IL-5的含量不具有顯著相關(guān)性,(r=0.48,P=0.1)；NP組中Eos計(jì)數(shù)和IL-5的含量具有顯著相關(guān)性,呈正相關(guān)(r=0.86,P=0.001)；NP伴AR組中Eos計(jì)數(shù)和IL-5的含量具有顯著相關(guān)性,呈正相關(guān)(r=0.84,P=0.001)。 5. 結(jié)論：單純性NP病人息肉組織Eos計(jì)數(shù)和IL-5表達(dá)均增加,合并AR時(shí)增加顯著；Eos浸潤鼻黏膜程度與IL-5表達(dá)密切相關(guān),IL-5可能參與NP的病理形成。
[Abstract]:Objective: To investigate the expression of IL-5 in polyp tissues of patients with nasal polyps, and whether there is a correlation with the Eos count.
Methods: combining with the history, nasal endoscopy, CT and laboratory examination the patients were divided into 3 groups: control group (13 cases), simple nasal polyps group (22 cases), NP with allergic rhinitis (allergic rhinitis, AR) group (20 cases). The control group of nasal septum deviation patients a portion of the middle turbinate hypertrophy. Polyps endoscopic removal, preservation of sections stained by.HE and Eos count, immunohistochemistry and enzyme-linked immunosorbent assay (enzyme-linked immuno sorbent assay, ELISA) single factor variance analysis was used to detect the IL-5 expression in tissue group expression of.Eos and IL-5 in the case of counting differences, Pearson correlation analysis study on the correlation between the expression of Eos and IL-5 by counting.
