d-δ-三烯生育酚對人晶狀體上皮SRA細(xì)胞增殖和凋亡的影響

【摘要】:目的研究d-δ-三烯生育酚對人晶狀體上皮SRA細(xì)胞增殖和凋亡的影響,探討后發(fā)性白內(nèi)障治療的新方法。方法將人晶狀體上皮SRA細(xì)胞分為對照組和實(shí)驗(yàn)組,對照組不加藥物干預(yù),實(shí)驗(yàn)組設(shè)3個(gè)藥物濃度,分別為30μmol·L-1、40μmol·L-1、50μmol·L-1的d-δ-三烯生育酚,藥物干預(yù)人晶狀體上皮SRA細(xì)胞24 h和48 h后,采用MTT法、Hoechst33258熒光染色和流式細(xì)胞儀分析人晶狀體上皮SRA細(xì)胞的增殖和凋亡情況。結(jié)果 d-δ-三烯生育酚呈濃度-時(shí)間依賴性抑制人晶狀體上皮SRA細(xì)胞的增殖,誘導(dǎo)其凋亡。30μmol·L-1、40μmol·L-1、50μmol·L-1的d-δ-三烯生育酚干預(yù)48 h對SRA細(xì)胞的增殖抑制率分別為(32.23±5.25)%、(54.17±1.63)%和(86.80±0.85)%,與對照組比較差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05);干預(yù)24 h增殖抑制率分別為(12.70±1.20)%、(19.53±0.95)%和(51.83±6.11)%,與對照組相比差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05)。干預(yù)24 h后,各實(shí)驗(yàn)組細(xì)胞晚期凋亡率Q2分別為(5.87±0.15)%、(11.37±0.85)%、(22.77±2.41)%,與對照組的(1.10±0.17)%相比,差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05);各實(shí)驗(yàn)組細(xì)胞早期凋亡率Q4分別為(3.57±0.32)%、(4.20±0.40)%、(8.00±0.50)%,與對照組的(2.53±0.15)%相比,差異均有統(tǒng)計(jì)學(xué)意義(均為P0.05)。結(jié)論 d-δ-三烯生育酚抑制人晶狀體上皮SRA細(xì)胞增殖,誘導(dǎo)其凋亡,具有抗晶狀體上皮細(xì)胞增殖的生物學(xué)作用,為治療后發(fā)性白內(nèi)障提供一種新思路。
[Abstract]:Objective to study the effect of d- Delta - three - tocopherol on the proliferation and apoptosis of human lens epithelial SRA cells and to explore a new method for the treatment of posterior - onset cataract. Methods human lens epithelial SRA cells were divided into control group and experimental group, control group without drug intervention, the experimental group with 3 concentration were 30 mol L-1 and 40 mol L-1, 50 mol - L-1 d- - three ene delta tocopherol, drug intervention of human lens epithelial SRA cells 24 h and 48 h, using the MTT method, Hoechst33258 staining and analysis of cell proliferation and apoptosis of human lens epithelial SRA cells by flow cytometry. Results d- Delta - three - ene tocopherol inhibited the proliferation of human lens epithelial SRA cells in a concentration - time dependent manner and induced apoptosis. 30 mol L-1, 40 mol, 50 mol - L-1 - L-1 - d- delta three ene 48 h intervention on SRA tocopherol cell proliferation inhibition rates were (32.23 + 5.25)% and (54.17 + 1.63)% and (86.80 + 0.85)%, compared with the control group were significant the difference (P0.05); intervention 24 h proliferation inhibition rate (12.70 + 1.20)% and (19.53 + 0.95)% and (51.83 + 6.11)%, compared with the control group, the differences were statistically significant (P0.05). After treatment for 24 h, the cells of each experimental group late apoptosis rate of Q2 was (5.87 + 0.15)% and (11.37 + 0.85)% and (22.77 + 2.41)%, and the control group (1.10 + 0.17)%, the differences were statistically significant (P0.05); the experimental group of early apoptosis the rate of Q4 were (3.57 + 0.32)% and (4.20 + 0.40)% and (8 + 0.50)%, and the control group (2.53 + 0.15)%, the differences were statistically significant (P0.05). Conclusion d- delta three ene tocopherol inhibits the proliferation and induces apoptosis of human lens epithelial SRA cells, which has a biological effect on the proliferation of lens epithelial cells, and provides a new idea for the treatment of posterior capsule opacification.
