MMP-1、TIMP-1及TGF-β1在風(fēng)濕性心臟病瓣膜組織中的表達及意義

【摘要】:目的探討基質(zhì)金屬蛋白酶-1(MMP-1)、基質(zhì)金屬蛋白酶組織抑制劑-1(TIMP-1)及轉(zhuǎn)化生長因子-β1(TGF-β1)在風(fēng)濕性心臟病(RHD)瓣膜組織中的表達及意義。方法采用Western blot及RT-PCR法檢測RHD瓣膜組織(實驗組,n=30)及無RHD瓣膜組織(對照組,n=15)中MMP-1、TIMP-1及TGF-β1蛋白與mRNA的表達,堿水解法檢測膠原代謝情況,Masson法進行膠原纖維染色并檢測膠原容積分?jǐn)?shù)(CVF)。分析MMP-1、TIMP-1及TGF-β1與CVF的相關(guān)性。結(jié)果 MMP-1及TGF-β1蛋白和mRNA在實驗組中的表達高于對照組,而TIMP-1蛋白和mRNA在實驗組中的表達低于對照組,差異有統(tǒng)計學(xué)意義(P0.05)。實驗組膠原含量及CVF均高于對照組(P0.05)。相關(guān)性分析顯示,MMP-1與TGF-β1呈正相關(guān),TIMP-1與TGF-β1呈負(fù)相關(guān),MMP-1及TGF-β1與CVF呈正相關(guān),TIMP-1與CVF呈負(fù)相關(guān)。結(jié)論 MMP-1、TGF-β1和TIMP-1參與了RHD瓣膜病變的纖維化進展。
[Abstract]:Aim to investigate the expression and significance of matrix metalloproteinase-1 (MMP-1), tissue inhibitor of metalloproteinase-1 (TIMP-1) and transforming growth factor-尾 1 (TGF- 尾 1) in (RHD) valve tissue of rheumatic heart disease. Methods the expression of MMP-1,TIMP-1, TGF- 尾 1 protein and mRNA in RHD valvular tissue (experimental group, n = 30) and non-RHD valvular tissue (n = 15) were detected by Western blot and RT-PCR methods, and collagen metabolism was detected by alkaline hydrolysis. Masson staining of collagen fibers and detection of collagen volume fraction (CVF). The correlation between MMP-1,TIMP-1, TGF- 尾 1 and CVF was analyzed. Results the expression of MMP-1, TGF- 尾 1 protein and mRNA in the experimental group was higher than that in the control group, while the expression of TIMP-1 protein and mRNA in the experimental group was lower than that in the control group (P0.05). The collagen content and CVF in the experimental group were higher than those in the control group (P0.05). The correlation analysis showed that MMP-1 was positively correlated with TGF- 尾 1, TIMP-1 was negatively correlated with TGF- 尾 1, MMP-1 and TGF- 尾 1 were positively correlated with CVF, and TIMP-1 was negatively correlated with CVF. Conclusion MMP-1,TGF- 尾 1 and TIMP-1 are involved in the progression of RHD valvular fibrosis.
