硫辛酸對快速起搏兔心房誘發(fā)的心房肌電與結(jié)構(gòu)重構(gòu)的抑制作用

【摘要】:目的:探討硫辛酸對快速起搏兔心房誘發(fā)的急性心房肌電重構(gòu)以及結(jié)構(gòu)重構(gòu)的抑制作用及其可能機制。方法:將45只健康新西蘭大耳白兔隨機分為3組:假手術(shù)組(n=10)、起搏組(RAP組,n=10)、起搏+硫辛酸注射組(LA組,n=10)。起搏頻率為600次·min~(-1),在起搏0、4、8、12 h分別進行程序刺激,測定心房有效不應(yīng)期(AERP),并計算AERP頻率適應(yīng)性。刺激結(jié)束后,取左心房組織,通過HE染色和Masson染色觀察心房組織結(jié)構(gòu)重構(gòu)情況;通過檢測丙二醛(MDA)、活性氧自由基(ROS)水平以及超氧化物歧化酶(SOD)活性觀察各組心房肌氧化應(yīng)激水平。結(jié)果:假手術(shù)組在不同基礎(chǔ)刺激周長(即S1S1周長)作用下,各時間點AERP無顯著性改變(P0.05);與假手術(shù)或LA組比較,RAP組AERP逐漸縮短(P0.05);RAP組起搏開始后8、12 h與起搏前(0 h)比較,AERP_(200~150)頻率適應(yīng)性不良(P0.05);LA組各時段頻率適應(yīng)性無明顯變化(P0.05)。心房組織HE染色:假手術(shù)組心房肌纖維排列整齊、有序;RAP組心房肌纖維排列紊亂,部分區(qū)域肌纖維溶解、斷裂;LA組心房肌的病理學改變輕于RAP組。心房組織Masson染色:假手術(shù)組心房肌纖維呈紅色,僅可見少量藍色的膠原纖維;RAP組心房肌間質(zhì)中膠原纖維顯著增多,分布紊亂;LA組心房肌間質(zhì)中的膠原纖維含量較RAP組明顯減少。氧化應(yīng)激指標:與假手術(shù)相比,RAP組心房組織中丙二醛、ROS含量明顯增加(P0.05),SOD活性下降(P0.05);與RAP組相比,LA組心房中丙二醛、ROS含量明顯減少,SOD活性升高(P0.05)。結(jié)論:在兔心房快速起搏模型中,LA干預(yù)能夠有效抑制心房肌電重構(gòu)和結(jié)構(gòu)重構(gòu)。
[Abstract]:Aim: to investigate the inhibitory effect and possible mechanism of lipoic acid on acute atrial electrical remodeling and structural remodeling induced by rapid pacing in rabbits. Methods: 45 healthy New Zealand white rabbits were randomly divided into 3 groups: sham operation group (n = 10), pacemaker group (LA) group (n = 10). The pacing frequency was 600 times / min ~ (-1). Program stimulation was carried out at 0 ~ (4) and 8 ~ (-1) h, respectively. The effective refractory period of atrium (AERP) was measured, and the frequency adaptability of AERP was calculated. At the end of the stimulation, the left atrial tissue was taken and the structural remodeling of the atrium was observed by HE staining and Masson staining. The levels of malondialdehyde (MDA), reactive oxygen free radical (Ros) and the activity of superoxide dismutase (SOD) were measured to observe the level of oxidative stress in atrial muscle of each group. Results: under the action of different basal stimulation perimeter (i.e. S1S1 circumference) in sham-operation group, Compared with sham operation or LA group, AERP of rap group decreased gradually 812 h after pacing began and 0 h before pacing. In HE staining of atrial tissue: atrial fibers were arranged neatly in sham-operation group, atrial fibers were disordered in ordered rap group, and some areas of muscle fibers were dissolved. The pathological changes of atrial muscle in LA group were lighter than those in RAP group. Masson staining of atrial tissue: in sham-operated group, atrial muscle fiber was red, only a small amount of blue collagen fiber was found in atrial interstitial tissue of rap group, and the content of collagen fiber in atrial interstitial of LA group was significantly lower than that in RAP group. Oxidative stress index: compared with the sham operation group, the content of malondialdehyde (MDA) Ros in atrial tissue increased significantly in the rap group, and the activity of SOD decreased significantly in the atrial tissue of the RAP group, and the content of malondialdehyde (Ros) in the atrium of the LA group decreased significantly compared with the control group (P 0.05). Conclusion: LA intervention can effectively inhibit atrial electrical remodeling and structural remodeling in rabbit atrial rapid pacing model.
