【摘要】：目的同步觀察心肌骨橋蛋白(OTN)、血清氨基末端B型利鈉肽前體(NT-proBNP)在過負(fù)荷心衰(HF)、過負(fù)荷心衰重置于正常負(fù)荷小鼠心臟的影響。方法選用雄性昆明種系小鼠C57BL/6野生型小鼠,分為Sham組、過負(fù)荷HF組、過負(fù)荷心衰重新置于正常負(fù)荷(HF-NL)組,觀察三組形態(tài)學(xué)改變、血清NT-proBNP濃度以及OPN mRNA表達(dá)。結(jié)果光鏡下Sham組小鼠心肌細(xì)胞結(jié)構(gòu)完整,肌纖維排列整齊;過負(fù)荷HF組小鼠心肌纖維間隙增寬,心肌細(xì)胞呈不同程度變性及肥大,部分心肌細(xì)胞呈局灶性壞死;HF-NL組與過負(fù)荷HF組比較,心肌纖維間隙增寬得到改善,心肌細(xì)胞變性、肥大以及局灶性壞死變輕;血清NT-proBNP濃度在Sham組、過負(fù)荷HF組、HF-NL組分別為(54.10±8.91)mg/L、(459.78±22.79)mg/L(與Sham組比較,P0.01)、(362.67±43.73)mg/L(與過負(fù)荷HF組比較,P0.01)。OPN mRNA表達(dá)在正常小鼠中的表達(dá)較弱〔(0.95±0.10)%〕;在過負(fù)荷HF組增強(qiáng)〔(3.38±0.12)%,與Sham組比較,P0.01〕,而在HF-NL組表達(dá)減輕〔(1.97±0.14)%,與過負(fù)荷HF組比較,P0.01〕。結(jié)論OPN參與過負(fù)荷心衰置于正常負(fù)荷后的心肌重塑過程,抑制OPN表達(dá)可能是減輕、減緩過負(fù)荷HF發(fā)生、發(fā)展的干預(yù)靶點(diǎn)之一。
[Abstract]:Objective to observe the effects of OTNN, NT-proBNPs, a precursor of serum amino-terminal B-type natriuretic peptide (NT-proBNPN), on the heart of mice with congestive heart failure (CHF) and its resetting to normal load mice. Methods male Kunming strain mice, C57BL / 6 wild-type mice, were used in this study. Sham group, HF group and HF-NL group were divided into three groups. Morphological changes, serum NT-proBNP concentration and OPN mRNA expression were observed in the three groups. Results under light microscope, the myocardial cell structure of Sham group was intact and the muscle fibers were arranged neatly. Compared with overload HF group, myocardial fiber gap widened, myocardial cells denatured and hypertrophic, and some myocardial cells showed focal necrosis. Compared with overload HF group, myocardial fiber gap widened and myocardial cell denaturation was improved in HF-NL group. Hypertrophy and focal necrosis became mild, serum NT-proBNP concentration in Sham group, The HF-NL group was 54.10 鹵8.91 mg / L (459.78 鹵22.79 mg / L vs Sham group, 362.67 鹵43.73 mg / L respectively) (compared with the overloaded HF group, the expression of P0.01 .OPN mRNA was weaker in normal mice than that in the overload HF group (0.95 鹵0.10); in the overload HF group, it increased 3.38 鹵0.12g / L; compared with that in the Sham group, P 0.01g / L; in the HF-NL group, the expression was reduced by 1.97 鹵0.14), and that in the overload HF group was increased by 3.38 鹵0.12g / L, compared with that in the Sham group, while in the HF-NL group, the expression of P0.01 / L was reduced to 1.97 鹵0.14. Conclusion OPN is involved in myocardial remodeling after overload in patients with heart failure after normal load. Inhibition of OPN expression may be one of the intervention targets to alleviate, slow down the occurrence and development of overload HF.
