【背景及目的】門脈高壓是使肝硬化病人死亡風險增高的重要因素之一。實驗性肝硬化大鼠肝臟中RhoA及Rho-kinase表達水平明顯高于正常大鼠，RhoA/Rho-kinase信號通道的激活導致血管平滑肌收縮并抑制其舒張，從而促進了門脈高壓的發(fā)生。相關研究報道，阿魏酸能夠降低膽固醇合成，后者合成過程的中間產物GGPP是RhoA活化的關鍵物質。本實驗旨在研究阿魏酸對肝硬化大鼠門脈壓力的作用，并探討相關分子機制�！痉椒ā坑媚懝芙Y扎方法制作肝硬化大鼠模型。實驗動物分為SHAM+NS、BDL+NS及BDL+SF三組，比較分析各組大鼠大體特征及血清肝功能指標。阿魏酸治療對肝臟纖維化程度的影響通過顯微鏡下觀察肝臟HE染色切片、透射電鏡觀察肝臟超微結構改變，以及免疫組化檢測肝臟a-SMA表達等綜合評定。免疫組化方法檢測各組大鼠肝臟中RhoA、Rho-kinase及eNOS的表達。分離、培養(yǎng)原代大鼠HSC，另購入人肝星狀細胞系LX-2，分別給予阿魏酸/阿魏酸+GGPP處理，流式細胞術檢測各處理因素作用下細胞凋亡率。原位肝灌注方法測定大鼠原位肝灌注壓力/肝內血管阻力，以及由低到高濃度梯度的甲氧明對各組大鼠肝...
