;撬釋χ匕Y急性胰腺炎大鼠Kupffer細胞P38MAPK信號轉導通路的影響

目的用;撬幔═aurine)對重癥急性胰腺炎(severe acute pancreatitis ,SAP)大鼠進行預處理,采用聯(lián)合酶原位灌注法分離枯否細胞(Kupffer cells,KCs),檢測KCs中P38-絲裂原蛋白激酶(P38 mitogen activated protein kinase, P38MAPK)的蛋白量和其活化水平以及核因子-κB(Nuclear factor-kappaB, NF-κB)的活性,探討;撬釋χ匕Y急性胰腺炎大鼠KCs內P38MAPK的影響以及對分泌促炎細胞因子的作用。方法48只雄性SD大鼠采用完全隨機法分為:①假手術對照組(sham operation,SO);②重癥急性胰腺炎組(SAP);③牛磺酸預處理組(Tau)。SAP模型通過胰膽管逆行注射5%;悄懰徕c誘導,造模前Tau組分別于24h和12h從尾靜脈內注入;撬幔300mmol/L,1.0ml /100g),其它組注入等量的生理鹽水。假手術或造模后分別于12h和24h處死動物,檢測其血清中AST、ALT和AMS含量;然后采用聯(lián)合酶原位灌注法分離KCs,采用蛋白免疫印跡法和免疫細胞化學...
