TGF-β1在BMSC移植可能對大鼠肝纖維及肝癌進程影響中的作用

目的建立骨髓間充質(zhì)干細胞體外培養(yǎng)及擴增體系的基礎(chǔ)上,探討不同時期移植BMSC、移植時間長短以及移植途徑不同對大鼠肝纖維化進程的影響:從TGF-β1的角度出發(fā),探討B(tài)MSC移植在肝纖維化進程中的作用機制以及可能在肝硬化后肝癌發(fā)生中的影響。方法1、應用密度梯度離心結(jié)合貼壁培養(yǎng)法分離純化雄性SD大鼠BMSC,分別向成骨細胞,成脂肪細胞誘導分化進行鑒定。2、在四氯化碳誘導大鼠肝纖維化模型的第六周、第八周,將來自雄性SD大鼠的BMSC以5×106/只的劑量經(jīng)尾靜脈注入同種雌性大鼠體內(nèi)。分別在BMSC移植4周及6周后處死各組大鼠,評價干預組與對照組的生存狀態(tài),并用Scheuer炎癥活動度評分法和Schmid M纖維化評分法評價肝臟病理情況。3、在四氯化碳誘導大鼠肝纖維化模型的第六周,將來自雄性SD大鼠的BMSC以5×106/只的劑量經(jīng)腹腔、經(jīng)尾靜脈及肝內(nèi)注射三種途徑移植入同種雌性大鼠體內(nèi)。BMSC移植6周后分別處死各組大鼠,評價各組的生存狀態(tài),異形細胞發(fā)生率,Scheuer炎癥活動度評分法和Schmid M纖維化評分法評價肝臟病理情況,免疫組化法檢測α-...
