敲除PEAK1破壞Src/FAK/Paxillin/Cortactin復(fù)合物維持的緊密連接加劇DSS誘發(fā)的小鼠腸炎

PEAK1是定位于細胞骨架actin等部位的非受體酪氨酸蛋白激酶,但是其對錨定在actin上的緊密連接以及緊密連接影響的潰瘍性結(jié)腸炎的調(diào)控機制仍不清楚。我們在PEAK1基因敲除小鼠中沒有觀察到自發(fā)性腸炎表型,但是在硫酸葡聚糖鈉誘導(dǎo)的潰瘍性結(jié)腸炎模型中,經(jīng)過體重、生存率、組織病理、炎癥因子等方面評估,首次證實了缺失PEAK1基因會加劇腸炎。根據(jù)結(jié)腸組織轉(zhuǎn)錄組測序結(jié)果的提示,我們采用免疫印跡實驗、免疫熒光實驗、免疫共沉淀等方法,在小鼠原代腸上皮細胞和腸上皮細胞系NCM460中探究了缺失PEAK1加劇腸炎的潛在分子機制。實驗結(jié)果表明,敲除PEAK1的腸炎小鼠結(jié)腸組織中促炎因子IL-6顯著上升,敲除PEAK1后腸上皮細胞中REG3β升高,敲低PEAK1后STAT3 Y705磷酸化明顯增加,因此我們提出解釋表型的第一個假說:PEAK1的缺失解除了REG3β對STAT3信號通路的抑制效應(yīng),釋放大量IL-6等炎癥促進因子,從而加重腸炎。在重點開展的緊密連接相關(guān)實驗中,敲除PEAK1顯著減少了AJs蛋白ZO-1和occludin的表達而對粘附連接標志物E-cadherin影響很小。我們對NCM460細...
