背景:咪喹莫特是Toll樣受體受體激動劑,可作為免疫佐劑,具有免疫調(diào)節(jié)作用,而潰瘍性結(jié)腸炎與免疫紊亂有密切相關(guān)性。目的:本研究旨在探討咪喹莫特(imiquimod,IMQ)對葡聚糖硫酸鈉(DSS)誘導(dǎo)的小鼠急性結(jié)腸炎的療效和可能機(jī)制。方法:將C57BL/6J小鼠隨機(jī)分為健康空白對照組、DSS模型組、地塞米松(dexa-methasone,DXM)組(1.5mg/kg)及咪喹莫特組(IMQ,30mg/kg),除正常組飲食自由外,其余各組飲用3%DSS溶液連續(xù)7天構(gòu)建實驗性UC小鼠模型,同時每天分別給小鼠腹腔注射相應(yīng)藥物治療,連續(xù)處理7天。治療期間記錄各組小鼠一般情況及死亡情況,在實驗第八天犧牲小鼠,比較各組小鼠結(jié)腸長度、脾重,及DAI評分,HE染色觀察小鼠結(jié)腸組織病理損傷變化,逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(RT-PCR)檢測腫瘤壞死因子-α(TNF-α)、白細(xì)胞介素-6(IL-6)、白細(xì)胞介素-10(IL-10)mRNA表達(dá)水平;酶聯(lián)免疫吸附法(ELISA)檢測小鼠血清炎癥因子 IL-6、TNF-α、IL-10 含量。結(jié)果:DSS成功誘發(fā)小鼠急性結(jié)腸炎,各實驗組小鼠有明顯腹瀉,黏液膿血便,體重減輕等急性結(jié)腸炎表現(xiàn),并且各實驗組小鼠均有不同程度死亡。IMQ能有效改善DSS誘導(dǎo)的急性結(jié)腸炎癥狀,并能減輕小鼠結(jié)腸炎癥嚴(yán)重程度。與DSS組相比,IMQ延緩了體重下降及結(jié)腸縮短(P0.01),增加了小鼠脾重(P0.05),下降疾病活動指數(shù)評分,降低結(jié)腸組織MPO活性(P0.01),并且炎癥因子IL-6、TNF-α表達(dá)水平降低(P0.01),IL-10表達(dá)水平升高(P0.01)。結(jié)論:咪喹莫特對DSS誘發(fā)的小鼠實驗性急性結(jié)腸炎有一定的治療效果,主要通過誘導(dǎo)特異性免疫應(yīng)答,增強(qiáng)機(jī)體免疫反應(yīng),降低炎癥反應(yīng)發(fā)揮作用。未來有望成為治療潰瘍性結(jié)腸炎的潛在藥物。
