【摘要】：目的探討小鼠腸道缺血再灌注損傷對(duì)體內(nèi)組織器官脂肪細(xì)胞因子chemerin的影響。方法無(wú)特定病原體級(jí)C57BL/6J小鼠16只,體質(zhì)量(25±2)g,適應(yīng)性飼養(yǎng)5 d后,按照隨機(jī)數(shù)字表尾數(shù)法分為腸道缺血再灌注組和假手術(shù)組,每組8只。腸道缺血再灌注組夾閉小鼠腸系膜上動(dòng)脈60 min,然后再灌注60 min造成腸道損傷,假手術(shù)組只開(kāi)腹不夾閉120 min。實(shí)驗(yàn)結(jié)束后,處死動(dòng)物取材檢測(cè)。應(yīng)用酶聯(lián)免疫吸附實(shí)驗(yàn)方法測(cè)定各組織器官chemerin的濃度變化,用Western blot方法檢測(cè)chemerin蛋白表達(dá)。結(jié)果酶聯(lián)免疫吸附實(shí)驗(yàn)結(jié)果顯示腸道缺血再灌注損傷后小鼠體內(nèi)各組織器官chemerin濃度顯著增加(P0.05),Western blot結(jié)果顯示chemerin蛋白表達(dá)顯著增加(P0.05)。結(jié)論腸道缺血再灌注損傷時(shí)小鼠各器官組織的脂肪細(xì)胞因子chemerin濃度和蛋白表達(dá)顯著增加。
[Abstract]:Objective to investigate the effect of intestinal ischemia reperfusion injury on adipocyte factor chemerin in tissues and organs of mice. Methods Sixteen C57BL/6J mice (25 鹵2 g) were randomly divided into intestinal ischemia-reperfusion group and sham-operation group according to the random number Mantissa method after 5 days of adaptive feeding. 8 mice in each group were divided into two groups: intestinal ischemia reperfusion group (n = 8) and sham operation group (n = 8). Intestinal injury was induced by occlusion of superior mesenteric artery for 60 min, and reperfusion for 60 min in the intestinal ischemia-reperfusion group, and no clamping was performed in the sham operation group for 120 min.. At the end of the experiment, the animals were sacrificed for examination. Enzyme linked immunosorbent assay (Elisa) was used to detect the concentration of chemerin in tissues and organs, and chemerin protein expression was detected by Western blot method. Results the results of enzyme-linked immunosorbent assay (Elisa) showed that the concentration of chemerin in tissues and organs of mice after intestinal ischemia-reperfusion injury was significantly increased (P0.05), Western blot results showed a significant increase in the expression of chemerin protein (P0.05). Conclusion the concentration of adipocyte factor chemerin and the expression of protein in various organs of mice were significantly increased during intestinal ischemia reperfusion injury.
