HBV感染者脂肪間充質(zhì)干細胞體外誘導為肝樣細胞

【摘要】:背景:有報道顯示,人脂肪間充質(zhì)干細胞在體外特定為環(huán)境下可誘導分化為具有白蛋白合成功能及分泌尿素功能的肝樣細胞。目的:探討乙型肝炎病毒感染者皮下脂肪組織間充質(zhì)干細胞體外向肝樣細胞分化的潛能。方法:膠原酶消化乙型肝炎病毒感染者脂肪組織,結(jié)合貼壁培養(yǎng)法獲得脂肪間充質(zhì)干細胞。采用三階段誘導法誘導脂肪間充質(zhì)干細胞向肝樣細胞分化,觀察其形態(tài)變化,免疫組織化學染色檢測肝細胞相關蛋白甲胎蛋白、白蛋白和細胞角蛋白18表達,糖原染色法檢測細胞糖原合成功能。結(jié)果與結(jié)論:脂肪間充質(zhì)干細胞經(jīng)3階段誘導培養(yǎng)后,大部分細胞成多角形,類肝細胞形態(tài);免疫組織化學染色結(jié)果顯示,誘導培養(yǎng)后脂肪間充質(zhì)干細胞白蛋白、細胞角蛋白18和甲胎蛋白染色陽性,且細胞白蛋白和細胞角蛋白18的表達隨著誘導培養(yǎng)時間的延長逐漸增加;誘導培養(yǎng)后的脂肪間充質(zhì)干細胞具有糖原合成功能,過碘酸雪夫試劑染色陽性。結(jié)果表明,乙型肝炎病毒感染者皮下脂肪組織間充質(zhì)干細胞經(jīng)成肝誘導培養(yǎng)后可分化為有功能的肝樣細胞。
[Abstract]:Background: it has been reported that human adipose mesenchymal stem cells can be induced to differentiate into hepatoid cells with albumin synthesis and urea secretion in vitro. Aim: to investigate the differentiation potential of subcutaneous adipose mesenchymal stem cells (MSCs) from hepatitis B virus infected patients into hepatocytes in vitro. Methods: adipose tissue of hepatitis B virus infected patients was digested by collagenase, and adipose mesenchymal stem cells were obtained by adherent culture. The differentiation of adipose mesenchymal stem cells into hepatocyte-like cells was induced by three-stage induction. The expression of hepatocyte associated protein alpha-fetoprotein, albumin and cytokeratin 18 was detected by immunohistochemical staining. Glycogen synthesis was detected by glycogen staining. Results and conclusion: after three stages of induction and culture, most of the adipose mesenchymal stem cells were polygonal and hepatocyte-like, and the results of immunohistochemical staining showed that the adipose mesenchymal stem cells had albumin in the cultured adipose mesenchymal stem cells. Cytokeratin 18 and alpha-fetoprotein were positive, and the expression of albumin and cytokeratin 18 gradually increased with the prolongation of culture time, and adipose mesenchymal stem cells had glycogen synthesis function. Scheffer's reagent of periodate was positive. The results showed that mesenchymal stem cells of subcutaneous adipose tissue induced by hepatitis B virus could differentiate into functional hepatocytes.
