雌激素對去勢大鼠肝脂肪變性影響及炎性機(jī)制

【摘要】:目的觀察雌激素對去勢大鼠肝脂肪變作用,探討雌激素肝保護(hù)作用及可能機(jī)制。方法 6周SD雌性大鼠隨機(jī)分為3組:假手術(shù)組、手術(shù)組和雌二醇組,術(shù)后1周,雌二醇組大鼠每天灌胃戊酸雌二醇(1 mg/kg)水溶液,其他組大鼠給予蒸餾水,連續(xù)12周。進(jìn)行胰島素耐量試驗(yàn),采用酶聯(lián)免疫法測定肝組織中腫瘤壞死因子-α(TNF-α)、白介素-6(IL-6)和單核細(xì)胞趨化蛋白-1(MCP-1)含量。蘇木素-伊紅染色觀察肝脂肪變性和炎癥狀況。結(jié)果與假手術(shù)組比較,手術(shù)組大鼠體重、肝重、內(nèi)臟脂肪重量[分別為(439.9±53.1)、(12.6±1.9)、(29.6±12.1)g]及血清谷丙轉(zhuǎn)氨酶(ALT)、谷草轉(zhuǎn)氨酶(AST)水平[分別為(57.8±5.4)、(128.4±10.1)U/L]明顯升高(P0.05);與手術(shù)組比較,雌二醇組大鼠體重、肝重、內(nèi)臟脂肪重量[分別為(387.3±40.7)、(10.3±2.1)、(24.1±6.5)g]及血清ALT、AST水平[分別為(44.5±4.8)、(98.2±9.6)U/L]明顯降低(P0.05)。手術(shù)組大鼠肝小葉結(jié)構(gòu)模糊,肝索排列紊亂,有廣泛的脂滴形成,小葉內(nèi)及匯管區(qū)可見炎癥細(xì)胞浸潤;雌二醇組大鼠肝小葉結(jié)構(gòu)仍欠清晰,脂滴明顯減少,且脂滴明顯變小;與手術(shù)組比較,雌二醇組大鼠肝脂肪變程度及小葉內(nèi)炎癥程度顯著減輕。與假手術(shù)組比較,手術(shù)組大鼠肝組織勻漿中TNF-α、IL-6、MCP-1含量[分別為(186.3±17.2)、(187.6±26.5)、(112.3±22.4)pg/m L]明顯升高(P0.05);與手術(shù)組比較,雌二醇組大鼠肝組織勻漿中TNF-α、IL-6、MCP-1含量[分別為(132.6±8.6)、(110.8±21.2)、(89.1±10.8)pg/m L]明顯降低(P0.05)。結(jié)論雌激素可減輕去勢大鼠肝脂肪變性程度,其機(jī)制可能與降低肝組織炎性因子表達(dá)有關(guān)。
[Abstract]:Objective to observe the effect of estrogen on hepatic adipogenesis in ovariectomized rats and to explore the protective effect and possible mechanism of estrogen on liver. Methods six weeks SD female rats were randomly divided into three groups: sham operation group, operation group and estradiol group. One week after operation, estradiol group rats were gavaged with estradiol valerate (1 mg/kg) solution daily, and other rats were given distilled water for 12 weeks. The contents of tumor necrosis factor- 偽 (TNF- 偽), interleukin-6 (IL-6) and monocyte chemoattractant protein-1 (MCP-1) in liver tissue were measured by enzyme linked immunosorbent assay (Elisa). Hepatic steatosis and inflammation were observed by hematoxylin-eosin staining. Results compared with sham operation group, the body weight, liver weight, visceral fat weight [(439.9 鹵53.1), (12.6 鹵1.9), (29.6 鹵12.1 g) g] and serum alanine aminotransferase (ALT),) transaminase (AST) levels [(57.8 鹵5.4), (128.4 鹵10.10) U / L] in the operation group were significantly higher than those in the operation group (P0.05), and the weight of the rats in the estradiol group was significantly higher than that in the operation group (P0.05). Liver weight, visceral fat weight (387.3 鹵40.7), (10.3 鹵2.1), (24.1 鹵6.5g) and serum ALT,AST level (44.5 鹵4.8), (98.2 鹵9.6U / L) were significantly decreased (P0.05). In the operation group, the hepatic lobule structure was blurred, the hepatic cord arranged disorderly, the lipid droplets were formed extensively, inflammatory cells were infiltrated in the lobules and in the catchment area, the structure of the hepatic lobules in the estradiol group was still unclear, the lipid droplets were obviously decreased, and the lipid droplets were obviously decreased. Compared with the operation group, the degree of hepatic fatty degeneration and intralobular inflammation in the estradiol group was significantly reduced. Compared with the sham operation group, the content of TNF- 偽 -IL-6 MCP-1 in the liver homogenate of the operation group [(186.3 鹵17.2), (187.6 鹵26.5), (112.3 鹵22.4 pg/m L, respectively)] was significantly higher (P0.05), and the content of TNF- 偽 -IL-6MCP-1 in the liver tissue homogenate of the Estradiol group was significantly lower than that in the control group [(132.6 鹵8.6), (110.8 鹵21.2), (89.1 鹵10.8) pg/m L] (P0.05). Conclusion estrogen can reduce the degree of hepatic steatosis in ovariectomized rats, and its mechanism may be related to the decrease of inflammatory factor expression in liver tissue.
