急性肝衰竭大鼠肝臟的蛋白質(zhì)組學研究

【摘要】:目的篩選急性藥物性肝衰竭大鼠肝臟組織的差異表達蛋白質(zhì)。方法將24只SD大鼠分為兩組,12只通過腹腔注射10g/L的D-氨基半乳糖建立大鼠急性肝衰竭模型(實驗組),12只腹腔注射生理鹽水作為對照組。提取兩組大鼠肝臟組織的蛋白質(zhì),定量后進行IEF和十二烷基磺酸鈉-聚丙烯酰胺凝膠電泳(SDS-PAGE)雙向凝膠電泳(2-DE)分離,通過軟件找到差異蛋白點并使用基質(zhì)輔助激光解吸電離飛行時間質(zhì)譜法(MALDI-TOF-MS)進行鑒定。結(jié)果成功鑒定出27個有效的差異蛋白質(zhì)點,其中實驗組較對照組上調(diào)15個,下調(diào)12個。結(jié)論急性藥物性肝衰竭模型大鼠肝臟酪蛋白激酶Iα(CKⅠα)、酪氨酸蛋白激酶(PTK)、增殖細胞核抗原(PCNA)等蛋白的表達較正常大鼠存在顯著差異。
[Abstract]:Objective to screen differentially expressed proteins in the liver tissues of rats with acute drug-induced liver failure. Methods Twenty-four Sprague-Dawley rats were divided into two groups: 12 rats were treated by intraperitoneal injection of D-galactosamine (10g/L) to establish acute hepatic failure model (experimental group: 12 rats were treated by intraperitoneal injection of normal saline as control group). Two groups of rat liver tissue proteins were extracted and separated by IEF and SDS-PAGE2-DEG after quantitative analysis. Differential protein sites were found by software and identified by matrix assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS). Results 27 effective differentially expressed protein spots were successfully identified, of which 15 were up-regulated and 12 down-regulated in the experimental group as compared with the control group.
