紫茉莉?qū)凭愿螕p傷大鼠ALT、AST、T-SOD影響的研究

        目的通過觀察紫茉莉根醇提物對酒精性肝損傷大鼠ALT、AST、T-SOD的影響,探討紫茉莉?qū)Υ笫缶凭愿螕p傷的保護作用。方法SD大鼠40只,雌雄各半,隨機分為3組:正常對照組10只、模型對照組及紫茉莉處理組各15只。正常組大鼠以蒸餾水灌胃,其余兩組大鼠均以白酒灌胃,連續(xù)灌胃80天,每天1次。第51天開始,正常組大鼠以蒸餾水灌胃,其余兩組大鼠均以相應藥物灌胃,連續(xù)灌胃30天,每天1次。第80天末次給藥后,禁食不禁水12h,摘眼球取血,處死大鼠取肝臟。測定血清中谷丙轉(zhuǎn)氨酶(ALT)活性、谷草轉(zhuǎn)氨酶(AST)活性及肝組織中總超氧化物歧化酶(T-SOD)活性;計算肝臟指數(shù);觀察肝組織病理變化。結(jié)果1、血清谷丙轉(zhuǎn)氨酶(ALT)檢測結(jié)果:與正常組比較,模型組和紫茉莉組大鼠血清中ALT活性均顯著升高(P <0.01),表明兩組大鼠肝臟均有不同程度損傷。與模型組比較,紫茉莉組大鼠血清中ALT活性顯著降低(P <0.01),表明紫茉莉組大鼠肝臟損傷程度有減輕。2、血清谷草轉(zhuǎn)氨酶(AST)檢測結(jié)果:與正常組比較,模型組和紫茉莉組大鼠血清中AST活性均顯著升高(P <0.01),,表明兩組大鼠肝臟均有不同程度損傷。與模型組比較,紫茉莉組大鼠血清中AST活性顯著降低(P <0.05),表明紫茉莉組大鼠肝臟損傷程度有減輕。3、肝組織中總超氧化物歧化酶(T-SOD)檢測結(jié)果:與正常組比較,模型組和紫茉莉組大鼠肝組織中T-SOD活性均顯著降低(P <0.01),表明兩組大鼠肝組織均有不同程度過氧化損傷。與模型組比較,紫茉莉組大鼠肝組織中T-SOD活性顯著升高(P <0.05),表明紫茉莉組大鼠肝組織過氧化損傷程度有減輕。4、肝臟指數(shù)檢測結(jié)果:與正常組比較,模型組和紫茉莉組大鼠肝臟指數(shù)均顯著增大(P <0.01),表明兩組大鼠肝臟均有不同程度腫大。與模型組比較,紫茉莉組大鼠肝臟指數(shù)顯著減。≒ <0.05),表明紫茉莉組大鼠肝臟腫大程度有減小。5、肝組織病理變化:正常組肝組織結(jié)構(gòu)正常;模型組肝細胞水樣變性(水腫)伴灶狀壞死;紫茉莉組與模型組比較,肝細胞水腫減輕,壞死減少,肝組織的形態(tài)結(jié)構(gòu)顯著改善。結(jié)論紫茉莉?qū)Υ笫缶凭愿螕p傷具有一定的保護作用,其機制可能與抑制過氧化反應,減輕酒精性肝病造成的肝細胞損傷有關。

    Objective: To observe the effects of Mirabilis Jalapa on ALT、AST、 T-SOD ofalcoholic liver injury rats and study the protecting effects on alcoholic liver injuryrats.Methods:40SD Rats were randomly divided to three groups, including model,Mirabilis Jalapa dealing and control groups. Hepatic injury models were establishedby intragastric administration of ethanol. The saline water and ethanol wereadministered intragastrically to the corresponding groups for80continuous days.From the51stday, the Mirabilis Jalapa was administered intragastriclly to accordinggroup for30continuous days. The blood sample and liver were collected in80thday. The concebtrations of aspartate aminotransferase(AST)、 alanineaminotransferase(ALT) and total superoxide dismutase(T-SOD) in serum wereassayed in according to the procedure described in kits. Then the hepatic indexes werecalculated and the pathological changes were observed.Results:1. The activities of ALT were improved in model and Mirabilis Jalapa groupcompared with control group. It indicated the liver injury model leading by ethanolwere established successfully. And the decline of the ALT level in Mirabilis Jalapagroup compared with model indicated the Mirabilis Jalapa can reduce the liver injuryleading by ethanol.2. The activities of AST were improved in model and Mirabilis Jalapa groupcompared with control group. It indicated the liver injury model leading by ethanolwere established successfully. And the decline of the AST level in Mirabilis Jalapagroup compared with model indicated the Mirabilis Jalapa can reduce the liver injuryleading by ethanol.3. The activities of T-SOD were decreased in model and Mirabilis Jalapa groupcompared with control group. It indicated the liver injury model leading by superoxide dismutase were established successfully. And the improve of the T-SODlevel in Mirabilis Jalapa group compared with model indicated the Mirabilis Jalapacan reduce the liver injury through inhibiting superoxide reaction.4. The hepatic index increased in model group and decreased in Mirabilis Jalapagroup compared with control. It indicated that the Mirabilis Jalapa can reduce thehypertrophy changing of liver leading by ethanol injury.5. The hydrophic degeneration and spotty necrosis of liver cells can be observed inmodel group and reduced in Mirabilis Jalapa group.Conclusion: The Mirabilis Jalapa can reduce the liver injury leading by ethanolthrough reducing the level of superoxide reaction to hepatic cells from ethanol.
