Kallistatin通過Akt-eNOS信號(hào)通路對(duì)肝星狀細(xì)胞氧化損傷的保護(hù)作用

【摘要】:為揭示kallistatin對(duì)肝星狀細(xì)胞氧化損傷的保護(hù)作用及相關(guān)分子機(jī)制,本文以過氧化氫和鐵過載為氧化模型,以大鼠原代肝星狀細(xì)胞及完全活化的人肝星狀細(xì)胞系LX-2為對(duì)象,研究了kallistatin在對(duì)肝星狀細(xì)胞活力、超氧化物、NADPH和Akt、e NOS等分子的影響。實(shí)驗(yàn)結(jié)果顯示,kallistatin既能避免肝星狀細(xì)胞受氧化損傷,還能修復(fù)氧化應(yīng)激造成的細(xì)胞損傷;其主要機(jī)制為kallistatin能清除細(xì)胞內(nèi)的超氧化物、降低細(xì)胞內(nèi)NADPH活力;另外,kallistatin還能激活A(yù)kt和e NOS等分子發(fā)揮抗氧化作用。本研究為加速肝纖維化藥物新靶點(diǎn)開發(fā)提供了理論依據(jù)。
[Abstract]:In order to reveal the protective effect of kallistatin on oxidative damage of hepatic stellate cells and its related molecular mechanisms, hydrogen peroxide and iron overload were used as oxidation models, and rat primary hepatic stellate cells and fully activated human hepatic stellate cell line LX-2 were used as subjects.The effects of kallistatin on the activity of hepatic stellate cells, superoxide NADPH and NOS were studied.The results showed that kallistatin could not only avoid oxidative damage of hepatic stellate cells, but also repair the oxidative stress, and its main mechanism was that kallistatin could scavenge the superoxide and decrease the activity of NADPH in the cells.In addition, kallistatin can also activate Akt, e NOS and other molecules to play an antioxidant role.This study provides a theoretical basis for accelerating the development of new drug targets for liver fibrosis.
